摘要:
目的:观察补肾活血汤对退变椎间盘中NLRP3/Caspase-1炎症小体的影响并探究补肾活血汤改善腰椎间盘退变的作用机制。方法:SD大鼠70只(雌雄各半),随机分为3组:Ⅰ组50只进行椎间盘退变模型建立,Ⅱ组10只作为空白组(A组),Ⅲ组10只作为假手术组(B组)。验证造模成功后,Ⅰ组取44只大鼠随机分为模型组8只(C组)、抑制剂组18只(D组,Caspase-1抑制剂灌胃)及中药组18只(E组,补肾活血汤灌胃)。各组于喂养2周后随机处死组内半数大鼠取出其L3/4至L5/6椎间盘组织,4周后将剩余大鼠全部处死取L3/4至L5/6椎间盘组织,样本行苏木精-伊红(HE)染色,Western Blot法检测NLRP3、Caspase-1蛋白表达水平,PCR检测NLRP3、Caspase-1基因表达水平,ELISA法测定IL-1β、TNF-α蛋白含量。结果:与空白组比较,模型组NLRP3、Caspase-1、IL-1β、TNF-α各项指标均显著高于空白组,差异有统计学意义(P<0.01); 与模型组比较,中药组NLRP3、Caspase-1、IL-1β、TNF-α各项指标均显著低于模型组,差异有统计学意义(P<0.01)。结论:补肾活血汤可以通过抑制椎间盘内NLRP3/Caspase-1炎症小体起到治疗或延缓椎间盘退变的作用。
Abstract:
Objective:To observe the efficacy of Bushen Huoxue decoction on NLRP3/Caspase-1 inflammasome in degenerative intervertebral discs and explore the mechanism of Bushen Huoxue decoction in improving lumbar intervertebral disc degeneration.Methods:70 SD rats(half male and half female)were randomly divided into three groups,50 rats in group Ⅰ were used to establish the intervertebral disc degeneration model,10 rats in group Ⅱ were used as the blank group(group A),and 10 rats in group Ⅲ were used as the sham operation group(group B).After verifying the successful modeling,44 rats in group Ⅰ were randomly divided into model group(group C,n=8),inhibitor group(group D,Caspase-1 inhibitor gavage,n=18),and traditional Chinese medicine group(group E,Bushen Huoxue decoction gavage,n=18).After feeding for 2 weeks,half of the rats in each group were killed,and their L3/4 to L5/6 intervertebral disc tissues were taken out.After 4 weeks,all the remaining rats were killed,and the L3/4 to L5/6 intervertebral disc tissues were taken out.The samples were stained with HE,the expression levels of NLRP3 and Caspase-1 proteins were detected by Western Blot,the expression levels of NLRP3 and Caspase-1 genes were detected by PCR,and the protein contents of IL-1β,TNF-α were detected by ELISA.Results:Compared with the blank group,all the indexes of NLRP3,Caspase-1,IL-1β,TNF-α in the model group were significantly higher than those in the blank group(P<0.01).Compared with the model group,all the indexes of NLRP3,Caspase-1,IL-1β,TNF-α in the traditional Chinese medicine group were significantly lower than those in the model group(P<0.01).Conclusion:Bushen Huoxue decoction can treat or delay intervertebral disc degeneration by inhibiting NLRP3/Caspase-1 inflammasome in the intervertebral disc.
