摘要:
目的:探究温肾宣痹方对大鼠膝骨关节炎(Osteoarthritis,OA)的作用效果及机制。方法:建立前交叉韧带切断的大鼠骨性关节炎动物模型,实验组给予温肾宣痹方灌胃8周,对照组、空白组给予纯净水灌胃8周。观察各组大鼠的国际骨关节炎研究协会(Osteoarthritis Research Society International,OARSI)评分以及软骨组织中MMP-1、MMP-13的基因的相对表达、滑膜组织中miR-146a的相对表达。结果:温肾宣痹方组的OARSI评分较对照组低、软骨组织中MMP-1、MMP-13的相对表达量较对照组低、滑膜组织中miR-146a的相对表达量较对照组高,差异有统计学意义(P<0.05)。结论:灌胃温肾宣痹方水煎剂对大鼠骨性关节炎具有一定的软骨保护效果,可延缓OA进程,机制与增加滑膜组织中miR-146a的表达相关。
Abstract:
To investigate the efficacy and potential mechanism of Wenshenxuanbi decoction(WSXBD)on knee osteoarthritis(OA)in rats. Methods:Rats model of anterior cruciate ligament(ACL)transection were established. The experimental group was given WSXBD for 8 weeks, while the control group and the blank group was given pure water for 8 weeks. The Osteoarthritis Research Society International(OARSI)score, the relative expression of MMP-1, MMP-13 in cartilage and miR-146a in synovium were observed.Results:The OARSI score of WSXBD was lower than that in control group, the relative expression of MMP-1 and MMP-13 was lower than that in control group, and the relative expression of miR-146a was higher than that in control Group. Conclusion:WSXBD has a protective efficacy on the cartilage of rat osteoarthritis, which can delay the progress of OA. The mechanism is related to the increase of miR-146a expression.
