摘要:
目的:探讨电针对不同造模时间下膝骨关节炎(KOA)模型兔滑膜组织中TLR4/NF-κB信号通路的影响。方法:采用改良Hulth法建立兔KOA模型,将44只家兔随机分为6组,其中空白组4只,假手术组、模型1组、模型2组、电针1组、电针2组各8只。空白组、假手术组、模型1组和模型2组不予治疗,电针1组及电针2组分别在造模后2周及4周给予电针治疗,每周治疗3次,共治疗4周。处死家兔后,取模型膝周滑膜组织,并应用实时荧光定量聚合酶链反应(PCR)及Western Blot法检测滑膜组织中TLR4、NF-κB、MyD88的mRNA及蛋白的表达。结果:PCR结果显示模型2组中TLR4、NF-κB及MyD88的mRNA表达量显著高于模型1组,电针治疗能够明显抑制TLR4、NF-κB及MyD88 的mRNA表达,差异有统计学意义(P<0.05)。Western Blot结果显示模型2组中TLR4、NF-κB及MyD88的蛋白表达量显著高于模型1组,电针治疗能够明显抑制TLR4、NF-κB及MyD88蛋白的表达,差异有统计学意义(P<0.05)。随着造模时间的延长,TLR4、NF-κB及MyD88的mRNA及蛋白表达水平呈上调的趋势。结论:电针治疗可以调控TLR4/NF-κB信号通路,抑制滑膜炎症反应,从而对膝骨关节炎起到治疗作用。
Abstract:
To investigate the efficacy of electroacupuncture on TLR4/NF-κB signaling pathway in synovial tissue of rabbits with knee osteoarthritis at different modeling time.Methods:44 rabbits were randomly divided into 6 groups: blank group, sham operation group, model 1 group, model 2 group, electroacupuncture group 1, electroacupuncture group 2, and 8 rabbits in each group. The blank group, the sham operation group, the model 1 group and the model 2 group were not treated. The electroacupuncture group 1 and the electroacupuncture 2 groups were given electroacupuncture treatment 2 weeks and 4 weeks after modeling. 3 times a week for 4 weeks. The expression of mRNA and protein of TLR4, NF-κB and MyD88 were detected by real-time PCR and Western Blot.Results:PCR results showed that mRNA expression levels of TLR4, NF-κB and MyD88 in model group 2 were significantly higher than that in model group 1. Electroacupuncture treatment significantly inhibited mRNA expression levels of TLR4, NF-κB and MyD88(P<0.05).Western Blot results showed that the protein expression levels of TLR4, NF-κB and MyD88 in model group 2 were significantly higher than that in model group 1. Electroacupuncture significantly inhibited the protein expression levels of TLR4, NF-κB and MyD88(P<0.05).The mRNA and protein levels of TLR4, NF-κB and MyD88 were up-regulated with the extension of modeling time.Conclusion:Electroacupuncture can regulate TLR4/NF-κB signaling pathway and inhibit synovial inflammatory reaction, thus playing a therapeutic role in knee osteoarthritis.
