摘要:
目的:探讨川芎嗪对大鼠岛状皮瓣缺血再灌注损伤的保护性作用。 方法:将112只SPF级SD大鼠按体重大小进行统一编号,随机分为假手术组(SO组)、对照组(IR组)、川芎嗪组(TMP组)和低分子右旋糖酐组(低右组),每组28只。每组动物再次随机分为2 h组、4 h组、6 h组、8 h组等亚组。以腹壁浅血管束为蒂,在大鼠腹部设计岛状皮瓣形成缺血再灌注模型,皮瓣组织取材及抽血检测,用硫代巴比妥酸TBA比色法和黄嘌呤氧化酶法分别测定大鼠皮瓣组织及血液中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性的变化指标。结果:(1)皮瓣内MDA与SOD活性比较,IR组的6 h和8 h亚组,皮瓣组织中内源性SOD活性均明显下降; TMP组和低右组的6h和8h亚组皮瓣组织中SOD活力均相应地有所提高,且明显高于同时间点IR组,差异有统计学意义(P<0.05); TMP组和低右组的6h和8h亚组皮瓣组织中MDA含量明显低于IR组(P<0.05); IR组间内,6h、8h亚组与2h、4h亚组相比较:SOD活力明显降低,差异有统计学意义(P<0.05),MDA含量升高(P<0.05)。(2)血清中MDA与SOD活性比较: IR组的各亚组血清中内源性SOD活性均明显下降,TMP组和低右组的6h和8h亚组血清中SOD活力均相应地有所提高,且明显高于同时间点IR组,差异有统计学意义(P<0.05),MDA含量明显低于IR组(P<0.05); IR组内,6h、8h亚组与2h、4h亚组相比,SOD活力差异有统计学意义(P<0.05); MDA含量升高(P<0.05)。结论:川芎嗪可降低皮瓣组织MDA水平,提高SOD活性,对游离皮瓣缺血再灌注损伤具有保护作用。
Abstract:
To study the protective effect of tetramethylpyrazine on rat island flap after ischemia-reperfusion injury.Methods:One hundred and twelve SPF SD rats were given serial numbers according to the weight and then randomly divided into sham operation group(SO), the control group(IR group), tetramethylpyrazine group(TMP group)and low molecular weight dextran group(low-right group ), 28 in each group. Animals in each group were randomly redivided into 2 h group, 4 h group, 6 h group, 8 h group and other subgroups. Ischemia-reperfusion model of abdominal island flap was created in the rats based on superficial epigastric vascular bundle. Flap tissues as well as blood samples were drawn for detection of malondialdehyde(MDA)content and superoxide dismutase(SOD)activity. Results:(1)MDA and SOD activity in the 6 h and 8 h IR groups as well as the endogenous SOD activity of flaps were significantly decreased. SOD activity of the flaps in 6h and 8h subgroups of TMP group and the low-right group were remarkably increased compared to IR group at the same time point(P<0.05). MDA content of the flaps in the TMP group and the low-right group(6h and 8h subgroups)was lower than the IR group(P<0.05). Within IR groups, SOD activity was significantly lower in 6h and 8h subgroups than that in 2h and 4h subgroups(P<0.05), while MDA content increased(P<0.05 ).(2)Endogenous SOD activity in serum was decreased in all subgroups of IR group, but it was increased significantly in 6h and 8h subgroups of TMP group and the low-right group compared to IR group at the same time point(P<0.05). MDA content was significantly lower in TMP group and the low-right group than the IR group(P<0.05). Among all IR groups, SOD activity was significantly decreased while MDA content increased in 6h and 8h subgroups compared with 2h and 4h subgroups(P<0.05). Conclusion:Tetramethylpyrazine can lower MDA level and increase SOD activity in flap tissues, thus has a protective effect on the free flap with ischemia-reperfusion injury.
