摘要:
目的:通过对去势大鼠整合素表达水平的观察,揭示金刚健骨片治疗骨质疏松症的作用机理。方法:将50只10月龄Wistar雌性大鼠随机分为5组:阿仑磷酸钠组、金刚健骨片组、骨松宝颗粒组、模型组、假手术组。治疗组及模型组行卵巢摘除术,假手术组切除相同重量的肠系膜。统一饲养13周造模成功后,治疗组分别以相应药物连续给药13周; 模型组及假手术组以生理盐水替代。治疗结束后处死动物取出骨组织,进行骨密度测定,并利用免疫组化法测定整合素αⅤβ6表达水平。结果:治疗组的骨密度、整合素αⅤβ6均较模型组有明显提高,差异有统计学意义(P<0.05); 金刚健骨片组与阿仑膦酸钠组对提高大鼠骨密度均高于骨松宝组(P<0.05),而两组比较差异无统计学意义(P>0.05); 治疗组的整合素αⅤβ6水平金刚健骨片组高于阿仑膦酸钠组及骨松宝组,差异有统计学意义(P<0.05)。结论:金刚健骨片能显著提高去势大鼠的骨密度、整合素αⅤβ6的表达水平,且较骨松宝颗粒疗效好,与阿仑磷酸钠治疗效果相当,为选出具有一定靶向性的治疗骨质疏松症的中药提供了新的思路。
Abstract:
To investigate the regulatory effect of Jingang Jiangu pill(JGJG)on integrin in ovariectomized osteoporosis rats. Methods:All 50 ovariectomized female rats, 10 months old, were randomly divided into 5 groups: alunlinsuanna group, Jingang Jiangu pill group, Gu Songbao granule group, model group, and the sham group. After ovariectomized, the rats were raised in the same environment for 13 weeks. Then treatment group were treated with different medicine for another 13 weeks, the model group and sham group were treated with saline instead. Bone mineral density(BMD)and the level of integrin αⅤβ6 were detected in each group after the treatment. Results:BMD, the level of integrin αⅤβ6 in treatment group were improved obviously than model group(P<0.05). The BMD in Jingang Jiangu pill group and alunlinsuanna group were improved obviously than Gu Songbao granule group(P<0.05), but there was no significant difference between the two groups(P>0.05). The level of integrin αvβ6 in Jingang Jiangu pill group was improved more than alunlinsuanna group and Gu Songbao granule group(P<0.05). Conclusion:Jingang Jiangu pill can improve BMD and level of integrin αvβ6 in ovariectomy rats, as well as alunlinsuanna, better than Gu Songbao granule, for providing a new Chinese medicine for elected certain targeted treatment of osteoporosis.
