摘要:
目的:研究土家药接骨膏促进骨折愈合的作用机理。方法:以新西兰兔建立左前肢桡骨骨折模型,实验分为接骨膏组、模型对照组及正常对照组,通过X线检测骨折愈合进程,血清钙磷检测,酶联免疫吸附试验(ELISA)法检测血清中IL-6、TNF-α、OC、PINP、FGF2、ALP及ACP5的浓度,苏木精-伊红(HE)染色检测骨折愈合进程,IHC检测骨折部位骨母细胞bFGF的表达,来研究接骨膏对新西兰兔骨折愈合的影响。结果:与模型对照组比较,接骨膏组的桡骨骨折线提前1周消失,血清钙峰值提前1周出现,钙磷乘积峰值显著升高,IL-6、TNF-α的下降速度更快、浓度更低(P<0.05或P<0.01),而血清中OC、PINP、FGF2、ALP的浓度更高(P<0.05或P<0.01),血清中ACP5浓度在造模后第7天达到峰值,随后呈快速下降趋势,显著提前骨痂、骨髓腔形成与贯通及骨钙化和血管生成的时间,骨母细胞bFGF的表达量更高。结论:接骨膏对新西兰兔桡骨骨折愈合具有促进作用。
Abstract:
Objective:To study the mechanism of traditional Chinese medicine(TCM)in promoting fracture healing.Methods:The New Zealand rabbits were used to establish the model of radial fracture of left forelimb, which were divided into Jiegu plaster group, model group and normal control group. The efficacy of Jiegu plaster on fracture healing of New Zealand rabbits were studied by X-ray examination of fracture healing process, serum calcium and phosphorus, ELISA examination of serum levels of IL-6, TNF-α, OC, PINP, FGF2, ALP and ACP5, HE staining examination of fracture healing process, IHC examination of bFGF expression of bone mother cells at fracture site.Results:Compared with the model control group, the radius fracture line disappeared 1 week earlier, the serum calcium peak value appeared 1 week earlier; the calcium-phosphorus product peak value increased significantly; IL-6 and TNF-α decreased more rapidly and at lower concentrations(P<0.05 or P<0.01), but OC, PINP, FGF2 and ALP in serum were higher(P<0.05 or P<0.01). The concentration of ACP5 in serum reached the peak at the first week after the model was established, and then decreased rapidly. The time of bone callus, bone marrow cavity formation and penetration, bone calcification and angiogenesis were advanced significantly, and the expression of bFGF was higher in osteoblasts.Conclusion:Jiegu ointment can promote the healing of radius fracture in New Zealand rabbits.
