摘要:
目的:探讨甘草甜素调控miR-4651表达对多发性骨髓瘤RPMI8226细胞增殖、凋亡的影响机制。方法:将多发性骨髓瘤RPMI8226细胞随机分为对照组、甘草甜素低、中、高浓度组、miR-4651组、miR-NC组、甘草甜素+anti-miR-4651组、甘草甜素+anti-miR-NC组; 用四甲基偶氮唑盐比色法(MTT)检测细胞增殖抑制率,流式细胞仪检测细胞凋亡,实时荧光定量聚合酶链式反应(RT-qPCR)检测miR-4651表达水平。结果:与对照组比较,甘草甜素低、中、高浓度组骨髓瘤RPMI8226细胞增殖抑制率、细胞凋亡率和miR-4651表达水平均逐渐升高(P<0.05),呈剂量依赖性。上调miR-4651表达后,骨髓瘤RPMI8226细胞增殖抑制率和细胞凋亡率均升高(P<0.05),而抑制miR-4651的表达则逆转了甘草甜素对骨髓瘤RPMI8226细胞增殖和凋亡的作用。结论:甘草甜素通过上调miR-4651表达可抑制多发性骨髓瘤RPMI8226细胞增殖,促进细胞凋亡。
Abstract:
Objective:To explore the mechanism of glycyrrhizin regulating the expression of miR-4651 on the proliferation and apoptosis of multiple myeloma RPMI8226 cells. Methods:Multiple myeloma RPMI8226 cells were randomly divided into control group, glycyrrhizin low, medium and high concentration group, miR-4651 group, miR-NC group, glycyrrhizin+anti-miR-4651 group and glycyrrhizin+anti-miR-NC group. Cell proliferation inhibition rate was detected by tetramethylazolium salt colorimetric method(MTT), apoptosis was detected by flow cytometry, real-time fluorescent quantitative PCR(RT-qPCR)was used to detect the expression level of miR-4651. Results:Compared with the control group, the proliferation inhibition rate, apoptosis rate and miR-4651 expression level of myeloma RPMI8226 cells in the low, medium and high concentration groups of glycyrrhizins were gradually increased in a dose-dependent manner(P<0.05). After up-regulating the expression of miR-4651, the proliferation inhibition rate and apoptosis rate of myeloma RPMI8226 cells were increased(P<0.05), while inhibiting the expression of miR-4651 reversed the efficacy of glycyrrhizin on the proliferation and apoptosis of myeloma RPMI8226 cells. Conclusion:Glycyrrhizin can inhibit the proliferation of multiple myeloma RPMI8226 cells and promote cell apoptosis by up-regulating the expression of miR-4651.
