摘要:
目的:观察补肝健腰方对腰椎间盘退变大鼠椎间盘TNF-α(Tumor Necrosis Factor-α)及NF-κB(Nuclear Factor-Kappa B)表达的影响。方法:SD大鼠75只,随机分为假手术组、模型组、腰痹通组、芬必得组、补肝健腰组,每组各15只,成功制备大鼠腰椎间盘退变模型,分别给予腰痹通胶囊、芬必得、补肝健腰方药物灌胃,检测各组干预前、干预第20天和第40天时椎间盘TNF-α及NF-κB表达,进行对比。结果:干预第20天和第40天,腰痹通组、芬必得组、补肝健腰组椎间盘TNF-α及NF-κB表达较模型组下降,差异有统计学意义(P<0.05),腰痹通组、芬必得组、补肝健腰组在干预第20天和第40天时椎间盘TNF-α及NF-κB表达水平,分别与本组干预前比较,差异有统计学意义(P<0.05),补肝健腰组在以上各时间点与腰痹通组、芬必得组比较,差异无统计学意义(P>0.05)。结论:补肝健腰方能降低腰椎间盘退变模型大鼠椎间盘TNF-α及NF-κB表达,这可能为该方可以防治椎间盘退变疾病的作用机理。
Abstract:
Abstract Objective: To observe the effects of Buganjianyao decoction on the expression of TNF-α and NF-κB in the intervertebral disc of rats with lumbar disc degeneration. Methods: All 75 SD rats were randomly divided into the sham-operated group, the model group, the Yaobitong group, the Ibuprofen sustained-release capsule group, and the Buganjianyao group, with 15 rats in each group. After the rat model of lumbar disc degeneration was prepared successfully, the groups were given Yaobitong capsule, Ibuprofen sustained-release capsule, Buganjianyao decoction by gavage, respectively. The expression of TNF-α and NF-κB in the intervertebral discs were observed and compared before, and at the 20th day and the 40th day after intervention. Results: At the 20th day and the 40th day after intervention, compared with the control group, the expression of TNF-α and NF-κB in the Yaobitong group, the Ibuprofen sustained-release capsule group and the Buganjianyao group were decreased significantly(P<0.05). There were significant differences when the expression of TNF-α and NF-κB on in the Yaobitong group, the Ibuprofen sustained-release capsule group, the Buganjianyao group at the 20th day, the 40th day after intervention compared with before intervention, respectively(P<0.05). There was no significant difference when the Buganjianyao group compared with the Yaobitong group, the Ibuprofen sustained-release capsule group at the above three times(P>0.05). Conclusions: Buganjianyao decoction could effectively lower the expression of TNF-α and NF-κB in intervertebral disc of rats with lumbar disc degeneration, which may be the mechanism of prevention and treatment of disc degeneration.
