摘要:
目的:模拟临床中老年女性绝经后合并骨关节炎的病理生理特点,建立膝骨关节炎合并骨质疏松症大鼠动物模型。方法:采用经典去卵巢法构建骨质疏松症模型后的大鼠加疲劳性跑台训练。18只6月龄雌性SD大鼠,采用抽签法随机均分为空白对照组(正常组)、单纯去卵巢组(骨质疏松组)和膝骨关节炎合并骨质疏松组(模型组),每组6只。单纯去卵巢组和模型组在无菌条件下摘除双侧卵巢后,单纯去卵巢组常规喂养,模型组术后恢复1周后行跑台疲劳性训练4周,空白组不作任何处理。5周后取右侧股骨、左侧膝关节、血清及尿液行组织学、力学及免疫学分析。结果:与空白组比较,单纯去卵巢组与模型组血清雌二醇浓度显著降低(P<0.01),最大载荷、断裂载荷降低(P<0.05); 与单纯去卵巢组比较,模型组大鼠尿液CTX-Ⅱ浓度明显升高(P<0.01),模型组大鼠关节软骨呈现轻度软骨损伤。结论:采用经典去卵巢法能构建骨质疏松症模型,并在此基础上使用动物跑台进行疲劳性运动可以造成膝关节劳损,从而成功建立符合女性中老年膝骨关节炎合并骨质疏松症的新模型。
Abstract:
Abstract Objective: To investigate the effect of red yeast rice on the expression of serum ALP and TRAP, TNF-α and RANK of bone tissue in ovariectomized osteoporosis rats. Methods: Forty female Wistar rats of 12 weeks old were randomly divided into the combination estrogen group, the red yeast rice group, the model group and the sham-operation group, with 10 rats in each group. The first three groups were treated with completely bilateral ovariectomy, while the sham-operation group were only treated to open the abdominal cavity without removing the ovaries. After 12 weeks of gavage administration, the animals of each group were sacrificed. The bone mineral density was measured by multi-spectral imaging system, the serum ALP and TRAP of rats in each group were measured by ELISA method, and the results of immunohistochemically staining image were quantitatively analyzed by Image Pro Plus. Results: The immunohistochemically expression level of TNF-α and RANK in the red yeast rice group were lower than that in the model group(P<0.05). Conclusion: Red yeast rice can improve the expression of serum ALP and reduce the expression of serum TRAP. It can improve the bone density of ovariectomized rats by reducing TNF-α and RANK levels. The mechanism is similar to that of estrogen, which may be related to the osteoblast and osteoclast activity.
