摘要:
目的:探讨正骨紫金丸对骨关节炎大鼠软骨组织的保护作用及其机制。方法:将Wistar大鼠随机分为对照组、模型组、阳性组、正骨紫金丸组、正骨紫金丸+3-甲基腺嘌呤组(正骨紫金丸+3-MA组),每组10只。苏木精-伊红(HE)染色观察大鼠关节软骨组织情况; TUNEL染色检测关节软骨组织的细胞凋亡率; ELISA检测血清中白细胞介素(IL)-6、肿瘤坏死因子(TNF)-α、诱导型一氧化氮合酶(iNOS)的含量; Kit试剂盒检测MDA含量和SOD活性; Western Blot检测软骨组织中PI3K/AKT/mTOR信号通路相关蛋白的表达。结果:与对照组相比,模型组大鼠关节软骨损伤明显,软骨细胞排列不规则,细胞聚集明显,软骨细胞凋亡率、促炎因子IL-6、TNF-α、iNOS、MDA含量及PI3K、AKT和mTOR蛋白的磷酸化升高,SOD活性、ULK1、Beclin1及LC3Ⅱ/Ⅰ的比值显著降低,差异有统计学意义(P<0.05); 阳性组和正骨紫金丸组中大鼠软骨组织损伤减轻,细胞排列整齐,细胞聚集明显减少,软骨细胞凋亡率、促炎因子IL-6、TNF-α、iNOS、MDA含量及PI3K、AKT和mTOR蛋白的磷酸化表达降低,SOD活性、ULK1、Beclin1及LC3Ⅱ/Ⅰ的比值显著升高,差异有统计学意义(P<0.05); 自噬抑制剂3-MA逆转了正骨紫金丸对关节软骨的保护作用,差异有统计学意义(P<0.05)。结论:正骨紫金丸能够抑制PI3K/AKT/mTOR信号通路,对骨关节炎大鼠的软骨组织发挥保护作用。
Abstract:
Objective:To investigate the protective effect of Zhenggu Zijin pill on cartilage tissue in osteoarthritis rats and elucidate its underlying mechanism.Methods:Wistar rats were randomly divided into five groups:control group,model group,positive group,Zhenggu Zijin pill group,and Zhenggu Zijin pill+3-methyladenine group(Zhenggu Zijin pill+3-MA),with 10 rats in each group.HE staining was used to observe the articular cartilage,while TUNEL staining was employed to detect the apoptosis rate of articular cartilage.The levels of serum interleukin(IL)-6,tumor necrosis factor(TNF-α),and inducible nitric oxide synthase(iNOS)were measured using ELISA.The content of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)were determined using a Kit.Western Blot analysis was performed to detect the expression of PI3K/AKT/mTOR signaling pathway-related proteins in cartilage.Results:Compared with the control group,the model group showed obvious damage to the articular cartilage,irregular arrangement of chondrocytes,and prominent cell clustering.The apoptosis rats of chondrocytes,as well as the levels of proinflammatory factors IL-6,TNF-α,iNOS,and MDA,were significantly increased in the model group.Additionally,the phosphorylation of PI3K,AKT,and mTOR proteins was elevated,while the SOD activity,ULK1,Beclin1,and the ratio of LC3Ⅱ/Ⅰ were decreased(P<0.05).In contrast,the cartilage tissue damage was reduced in the positive group and Zhenggu Zijin pill group,with orderly arrangement of cells and reduced cell aggregation.Moreover,the apoptosis rats of chondrocytes,levels of proinflammatory factors,and the phosphorylation of PI3K,AKT,and mTOR proteins were decreased,while the SOD activity,ULK1,Beclin1,and the ratio of LC3Ⅱ/Ⅰ were increased(P<0.05).Notably,the protective effect of Zhenggu Zijin pill on articular cartilage was reversed by the autophagy inhibitor 3-MA(P<0.05).Conclusion:Zhenggu Zijin pill exhibits a protective role in cartilage tissue of osteoarthritis rats by inhibiting the PI3K/AKT/mTOR signaling pathway.These findings provide valuable insights for the potential therapeutic application of Zhenggu Zijin pill in osteoarthritis treatment.
