摘要:
目的:比较身痛逐瘀汤对不同静水压下人退变髓核细胞形态及凋亡的影响。方法:选取2只雄性新西兰兔,体质量分别为3.2 kg和3.8 kg,按“兔表面积/人的表面积=兔的给药量/人的给药量”的换算方法,计算出兔的中药颗粒给药剂量,将颗粒溶于水后灌胃,灌胃结束后腹主动脉采血,制备身痛逐瘀汤含药血清。将人髓核细胞分为两组:单纯压力组(不加任何药物)和中药组(15%身痛逐瘀汤含药血清),比较两组髓核细胞在0.3,1.0,3.0 MPa压力下作用2,4,6 h后的细胞形态变化,采用TEM观察两组髓核细胞超微结构的变化及差异; 采用Annexin V-FITC/ PI检验两组髓核细胞的凋亡状况。结果:两组髓核细胞在0.3,1.0,3.0 MPa静水压下作用后突起均有断裂,断裂程度随压力增加而增加。同一压力和作用时间下,中药组比压力组髓核细胞形态及超微结构保存更完整,差异有统计学意义(P<0.05); 中药组和压力组髓核细胞凋亡率随压力和作用时间的增加而增加。同一压力及作用时间下中药组髓核细胞凋亡率比单纯压力组降低,差异有统计学意义(P<0.05)。3.0 MPa下两组髓核细胞的凋亡率显著增加,差异有统计学意义(P<0.05)。结论:身痛逐瘀汤含药血清在低静水压(0.3,1.0 MPa)下能一定程度上减少髓核细胞凋亡,而高静水压下(3.0 MPa)效果有限,说明本方对早期椎间盘退变有防治作用。
Abstract:
Objective:To compare the efficacy of Shentong Zhuyu decoction on the morphology and apoptosis of human degenerative nucleus pulposus cells under different hydrostatic pressure.Methods:Two male New Zealand rabbits with body mass of 3.2 kg and 3.8 kg were selected.According to the conversion method of “rabbit surface area/human surface area=rabbit dose/human dose”,the dosage of traditional chinese medicine granules was calculated.The granules were dissolved in water and then administered by gavage.After gavage,blood was collected from abdominal aorta to prepare the medicated serum of Shentong Zhuyu decoction.Human nucleus pulposus cells were divided into two groups:simple pressure group(without any drugs)and traditional Chinese medicine group(15% Shentong Zhuyu decoction containing serum).The morphological changes of nucleus pulposus cells in the two groups were compared after 2,4 and 6 h under the pressure of 0.3,1.0 and 3.0 MPa.The changes and differences of ultrastructure of nucleus pulposus cells in the two groups were observed by TEM; Annexin V-FITC/PI was used to test the apoptosis of nucleus pulposus cells in the two groups.Results:The processes of nucleus pulposus cells in both groups were broken under hydrostatic pressure of 0.3,1.0 and 3.0 MPa,and the degree of fracture increased with the increase of pressure.Under the same pressure and action time,the morphology and ultrastructure of nucleus pulposus cells in traditional chinese medicine group were more intact than those in pressure group(P<0.05); The apoptosis rate of nucleus pulposus cells in traditional chinese medicine group and pressure group increased with the increase of pressure and action time.Under the same pressure and action time,the apoptosis rate of nucleus pulposus cells in traditional chinese medicine group was lower than that in simple pressure group(P<0.05).The apoptosis rate of nucleus pulposus cells in the two groups increased significantly at 3.0 MPa.Conclusion:The medicated serum of Shentong Zhuyu decoction can reduce the apoptosis of nucleus pulposus cells to a certain extent under low hydrostatic pressure(0.3,1.0 MPa),but the efficacy is limited under high hydrostatic pressure(3.0 MPa),indicating that this prescription has a preventive and therapeutic efficacy on early intervertebral disc degeneration.
