摘要:
目的:探讨活骨灌注液对激素刺激骨髓间充质干细胞(BMSCs)后增殖和凋亡的影响。方法:全骨髓贴壁细胞培养法分离获得大鼠BMSCs并用流式细胞仪鉴定,设置梯度浓度活骨灌注液后通过CCK8法筛选作用终浓度。后将BMSCs分为三组:正常组(常规培养BMSCs)、激素组(BMSCs+1 μmol/L地塞米松)和药物组(激素组基础上+活骨灌注液)。采用MTT法检测细胞增殖、流式细胞仪检测细胞凋亡及Western Blot法检测相关凋亡蛋白的表达。结果:流式细胞仪检测CD29、CD90、CD45抗体后明确提取细胞为BMSCs,CCK8筛选出2-6的活骨灌注液为作用终浓度。地塞米松刺激后可抑制BMSCs的增殖并促进其凋亡,但活骨灌注液作用后,可缓解地塞米松的作用(相关检测结果显示P<0.05,差异有统计学意义)。另外Western Blot结果显示,激素组中Bax和Cytochrome C蛋白表达明显升高,Bcl-2表达明显降低,差异有统计学意义(P<0.05); 而经活骨灌注液处理后,Bax和Cytochrome C蛋白表达明显降低,Bcl-2表达明显升高,差异有统计学意义(P<0.05)。结论:活骨灌注液可促进地塞米松作用后BMSCs的增殖并抑制其凋亡。
Abstract:
Objective:To investigate the efficacy of Huogu perfusion on the proliferation and apoptosis of bone marrow mesenchymal stem cells(BMSCs)stimulated by dexamethasone.Methods:The whole bone marrow adherent cell culture method was used to isolate rat BMSCs and were identified by flow cytometry.After setting the gradient concentration of Huogu perfusion,the final concentration was screened by the CCK8 method; then the BMSCs were divided into three groups:normal group(conventional culture BMSCs),dexamethasone group(BMSCs+1 μmol/L dexamethasone),and drug group(based on dexamethasone group + Huogu perfusion).MTT method was used to detect cell proliferation.Flow cytometry was used to detect cell apoptosis.The Western Blot method was used to detect the expression of related apoptotic proteins.Results:After the detection of CD29,CD90,and CD45 antibodies by flow cytometry,it was clear that the cells were BMSCs,and was screened as the final concentration by CCK8 2-6 Huogu perfusion.Dexamethasone can inhibit the proliferation of BMSCs and promote their apoptosis after stimulation,but after the effect of Huogu perfusion,it can effectively alleviate the efficacy of dexamethasone,and the difference was statistically significant(P<0.05).Besides,Western Blot results showed that the expression of Bax and Cytochrome C protein in the dexamethasone group was significantly increased; the expression of Bcl-2 was significantly reduced,and the difference was statistically significant(P<0.05).After the treatment with Huogu perfusion,Bax and Cytochrome C protein expression were significantly reduced; Bcl-2 expression was significantly increased,and the difference was statistically significant(P<0.05).Conclusion:Huogu perfusion can effectively promote the proliferation of BMSCs and inhibit their apoptosis after dexamethasone.
