摘要:
目的:探索生骨再造丸对激素性股骨头坏死(SANFH)兔的干预作用及相关机制。方法:50只新西兰大白兔按随机数字表法分为空白组10只和实验组40只,实验组采用大肠杆菌脂多糖(LPS)联合甲泼尼龙琥珀酸钠造模法建立SANFH兔模型32只,按随机数字表法分为模型组10只,仙灵骨葆组11只,生骨再造丸组11只。仙灵骨葆组和生骨再造丸组分别以0.4 g/kg仙灵骨葆和0.6 g/kg生骨再造丸灌胃,28 d后取出两侧股骨,提取骨髓组织中骨髓间充质干细胞(BMSCs)、成骨细胞(OB)及脂肪细胞,qRT-PCR检测miRNA-708-3p和miRNA-708-5p表达水平。结果:模型组BMSCs、OB和脂肪细胞中的miRNA-708-3p、miRNA-708-5p与空白组相比表达水平均有升高,差异有统计学意义(P<0.01); 仙灵骨葆组和生骨再造丸组BMSCs、OB中miRNA-708-3p、miRNA-708-5p与模型组相比表达水平降低,差异有统计学意义(P<0.01)。结论:SANFH兔骨髓组织中miRNA-708-3p与miRNA-708-5p的表达升高是SANFH发生的机制之一,生骨再造丸抑制SANFH兔BMSCs、OB中的miRNA-708-3p与miRNA-708-5p表达水平是防治早期SANFH的分子机制。
Abstract:
To explore the interventional efficacy of Shenggu Zaizao pills on steroid-induced osteonecrosis of the femoral head(SANFH)rabbits and related mechanisms.Methods: 50 New Zealand white rabbits were divided into a blank group of 10 and an experimental group of 40 according to the random number table.32 SANFH rabbit models used Escherichia coli lipopolysaccharide(LPS)combined with methylprednisolone sodium succinate were built in the experimental group.They were divided into model group of 10, Xianling Gubao group of 11, Shenggu Zaizao pills group of 11 by random number table method.Xianling Gubao group and Shenggu Zaizao pills group were given 0.4 g/kg Xianling Gubao and 0.6 g/kg Shenggu Zaizao pills by gavage respectively.After 28 d, both femurs were taken out, and bone marrow mesenchymal stem cells(BMSCs), osteoblasts(OB)and fat cells were extracted from the bone marrow tissue.The expression levels of miRNA-708-3p and miRNA-708-5p were detected by qRT-PCR.Results:The expression levels of miRNA-708-3p and miRNA-708-5p in BMSCs, OB and adipocytes of the model group were increased compared with the blank group(P<0.01).The expression level of miRNA-708-3p and miRNA-708-5p in OB and BMSCs of Xianling Gubao group and Shenggu Zaizao pills group were lower than that of the model group(P<0.01).Conclusion:The increased expression of miRNA-708-3p and miRNA-708-5p in SANFH rabbit bone marrow tissue is one of the mechanisms of SANFH.The expression levels of miRNA-708-3p and miRNA-708-5p in SANFH rabbit BMSCs and OB inhabited by Shengu Zaizao pills is a molecular mechanism for the prevention and treatment of early SANFH.
