摘要:
目的:观察加味阳和汤对膝骨关节炎兔关节软骨骨形态发生蛋白-2(BMP2)及Sry相关组蛋白9(Sox9)表达的影响。方法:将健康新西兰大白兔随机分为正常组、模型组和实验组。除正常组外,其余2组动物均按照改良赫尔特法(Hulth法)制作膝骨关节炎模型后,分组处理,4,6,8周后取材,制作标本,行免疫组化染色观察Ⅱ型胶原(Col Ⅱ)的变化、 实时聚合酶链反应(PCR)监测软骨组织中BMP2和Sox9的表达情况、TUNEL监测软骨组织中软骨细胞凋亡情况。结果:正常组Ⅱ型胶原的表达量最高,模型组表达量最少,骨性关节炎时间越长阳性染色越浅,同一时间段内实验组染色较好,且分布较均匀; BMP2在正常软骨中少量表达,而在骨性关节炎软骨中有大量表达,骨性关节炎时间越久,BMP2的表达量越多,同一时间段内实验组的BMP2的表达量比模型组低; 而Sox9在正常组中有大量表达,而在模型组中表达量明显减少,在同一时间段内实验组均较模型组的表达量增高。正常组软骨中凋亡的软骨细胞数量较少,而在模型组中同一时间段内软骨细胞凋亡数量较正常组明显增加,骨性关节炎时间越久凋亡细胞数量越多,实验组经治疗后细胞调亡数量减少。结论:加味阳和汤能够下调膝关节中BMP2的表达量,上调Sox9的表达量,从而减缓骨性关节炎软骨退变,促进Ⅱ型胶原的表达,进而减少软骨细胞的凋亡。
Abstract:
To observe the effect of Jiawei Yanghe decoction on the expression of bone morphogenetic protein-2(BMP2)and Sry-related HMG-box gene 9(Sox9)in articular cartilage of rabbits with knee osteoarthritis.Methods:Healthy New Zealand white rabbits were randomly divided into normal group,model group and experimental group.In addition to the normal group,knee osteoarthritis model was made according to the modified Hulth method,after 4,6 and 8 weeks of treatment,samples were taken and specimens were made,the changes of typeⅡcollagen(Col Ⅱ)were observed by immunohistochemistry,the expression of BMP2 and Sox9 in cartilage was monitored by PCR,and the apoptosis of chondrocytes was monitored by TUNEL.Results:The expression of type Ⅱ collagen was the highest in the normal group,the least in the model group,the longer the osteoarthritis time,the lighter the positive staining,the staining in the experimental group was better in the same period,and more evenly distributed.BMP2 was scarcely present in normal articular cartilage,but there was a lot of expression in osteoarthritis,the longer the osteoarthritis time,the more BMP2 expression,and the expression of BMP2 in the experimental group was lower than that in the model group in the same period.However,Sox9 was largely expressed in normal articular cartilage,but significantly reduced in the model group,and amount of expression in the experimental group was higher than that in the model group in the same period.The number of apoptotic chondrocytes in the cartilage of the normal group was little,but the number of apoptotic chondrocytes in the same period of time in the model group was significantly higher than that in the normal group,the number of apoptosis increased with osteodystrophy time,and the number of apoptosis decreased in the experimental group after treatment.Conclusion:Jiawei Yanghe decoction can decrease the expression of BMP2 and up regulate the expression of Sox9 in knee joint,which can slow down the degeneration of osteoarthritis cartilage,promote the expression of type Ⅱ collagen,and reduce the apoptosis of chondrocytes.
