摘要:
目的:探讨不同浓度丹参酮ⅡA对软骨细胞Ⅱ型胶原及Wnt/β-catenin信号通路的影响。方法:提取新生SD大鼠原代软骨细胞,并通过Ⅱ型胶原蛋白免疫荧光进行鉴定,取P1代细胞进行实验,共设5组,其中1组设为空白对照组,另外4组分别用6.25 μmol/L,12.5 μmol/L,25 μmol/L及50 μmol/L浓度丹参酮ⅡA对其进行干预,24 h后通过蛋白印迹分析法(Western Blot)检测软骨细胞Ⅱ型胶原及β-catenin蛋白的表达情况。结果:随着丹参酮ⅡA浓度的增加,Ⅱ型胶原蛋白表达增加,12.5 μmol/L,25 μmol/L及50 μmol/L丹参酮ⅡA组Ⅱ型胶原蛋白量均较对照组增高(P<0.05),且Ⅱ型胶原蛋白量与丹参酮ⅡA浓度呈正相关(r=0.949,P<0.001),各丹参酮ⅡA组β-catenin蛋白量均较对照组降低(P<0.05),且β-catenin蛋白量与丹参酮ⅡA浓度呈负相关(r=-0.949,P<0.001)。结论:丹参酮ⅡA可抑制Wnt/β-catenin信号通路,促进软骨细胞Ⅱ型胶原表达,延缓软骨细胞退变。
Abstract:
Abstract Objective:To investigate the effect of Tanshinone ⅡA(TAN ⅡA)on collagen Ⅱ and Wnt/β-catenin signal pathway of rat chondrocytes in vitro. Methods: SD rat articular chondrocytes were isolated, and immunofluorescence was performed to identify the cells. A total of 5 groups were set up, 1 of which was a blank control group, and the other 4 groups were treated with different concentrations of TAN ⅡA(6.25 μmol/L, 12.5 μmol/L, 25 μmol/L and 50 μmol/L). The expression of β-catenin and collagen Ⅱ were examined by Western-Blot after 24 h.Results: The groups of 12.5 μmol/L, 25 μmol/L, 50 μmol/L concentration of TAN ⅡA expressed higher collagen Ⅱ than the control group(P<0.05). The expression of collagen Ⅱ was increased as the concentration of TAN ⅡA had a rising trend(r=0.949, P<0.001). All the groups treated with TAN ⅡA expressed lower β-catenin than the control group(P<0.05). The expression of β-catenin was decreased as the concentration of TAN ⅡA had a rising trend(r=-0.949,P<0.001). Conclusion: TAN ⅡA can inhibit the Wnt/β-catenin signaling pathway, promote the expression of type Ⅱ collagen in chondrocytes, and delay the degeneration of chondrocytes.
