摘要:
目的:观察通络止痛方对人膝骨关节炎关节液白细胞介素-1β(IL-1β)、透明质酸(HA)及一氧化氮(NO)含量的影响。方法:行关节镜手术患者,在术中抽取关节腔内积液,离心后取上清,加入不同浓度通络止痛方干预,分为高浓度组(2.5 mg/mL)、中浓度组(0.25 mg/mL)、低浓度组(0.025 mg/mL)和对照组(磷酸缓冲盐溶液,PBS)。应用酶联免疫吸附法(ELISA)检测各浓度组对关节液干预后IL-1β,HA及NO含量的变化。结果:高、中、低浓度组较对照组关节液中IL-1β及NO含量明显降低(P＜0.05),且IL-1β及NO的浓度与通络止痛方的浓度呈正相关(IL-1β,r=-0.29,P＜0.05; NO,r=-0.51,P＜0.05)。高、中、低浓度组较对照组关节液中HA含量明显升高(P＜0.05),且HA的浓度与通络止痛方的浓度呈正相关(r=0.59,P＜0.05)。结论:通络止痛方对关节液中IL-1β及NO有抑制作用,浓度越高抑制程度越大,通络止痛方对关节液中HA有促进作用,浓度越高促进程度越大。
Abstract:
Objective:To observe the effect of Tongluozhitong recipe on the expression of interleukin-1β(IL-1β),hyaluronic acid(HA)and nitric oxide(NO)in synovial fluid of human knee osteoarthritis.Methods:Patients who underwent arthroscopic surgery in our hospital were taken.The intra-articular effusion was extracted during the surgery,from which the supernatant was taken after centrifugation,and different concentration of Tongluozhitong recipe were added.The mixture were divided into the high concentration group(2.5 mg/mL),the middle concentration group(0.25 mg/mL),the low concentration group(0.025 mg/mL)and the control group(PBS).The level changes of IL-1β,HA and NO in synovial fluid were observed by enzyme linked immunosorbent assay(ELISA)after treated with different concentration groups.Results:The levels of IL-1β and NO in synovial fluid in the high,middle and low concentration group were decreased significantly compared with the control group(P＜0.05),and the concentration of IL-1β and NO were positively correlated with the concentration of Tongluozhitong recipe(IL-1β,r=-0.29,P＜0.05; NO,r=-0.51,P＜0.05).The levels of HA in the high,middle and low concentration group were significantly increased compared with the control group(P＜0.05),and the concentration of HA were positively correlated with the concentration of Tongluozhitong recipe(r=0.59,P＜0.05).Conclusion:The Tongluozhitong recipe has inhibitory effect on the expression of IL-1β and NO in synovial fluid,the higher concentration with greater degree of inhibition.While,which has synergistic effect on the expression of HA in synovial fluid,and the higher concentration with greater degree of promotion.
