摘要:
目的:观察不同剂量红花注射液对兔脊髓慢性损伤后的治疗作用。方法:将60只健康青紫蓝家兔随机分为低剂量组(A组)、中剂量组(B组)、高剂量组(C组)、盐水对照组(D组),正常饲养1个月,采用螺钉植入法建立慢性脊髓损伤动物模型。造模成功后,实验组各组分别通过耳缘静脉注射不同剂量的红花注射液,连续注射2周,对照组注射生理盐水。治疗后每组分别于术前1个月、术后6个月、给药后2周随机抽取5只家兔,以伤处为中心取出1.0 cm的脊髓组织,通过免疫组化进行MMP-2检测。同时对每组随机抽取的5只家兔进行心脏采血,通过ELISA法检测血清中MMP-12的表达变化,并于不同时间点分别通过斜板试验、改良Tarlov评分评价家兔行为学变化。结果:1)各组在造模后MMP-2,MMP-12的表达以及行为学评价较造模前比较,差异有统计学意义(P<0.05)。2)给药后A,B,C组MMP-2,MMP-12表达及行为学评价较D组比较,差异有统计学意义(P<0.05),中剂量组MMP-2,MMP-12的表达以及行为学评价较高,与低剂量组比较差异有统计学意义(P<0.05)。结论:中剂量红花注射液对治疗脊髓慢性损伤较其它剂量效果显著提高,此剂量可作为红花注射液治疗脊髓慢性损伤的参考剂量。
Abstract:
Objective:To observe the therapeutic effect of different doses with Honghua injection for treating the spinal cord injury of rabbits.Methods:All 60 rabbits were randomly divided into group A(low dose),group B(middle dose),group C(high dose),and group D(control group),with normal feeding for one month.The animal models of chronic spinal cord injury were established by screw implantation.Then the animals of group A,B,and C were intravenous injected with Honghua injection,and group D were intravenous injected with saline,once a day,for 2 weeks.Five rabbits in each group were randomly selected at 1 month before surgery,6 month after surgery,and 2 weeks after treatment.The 1.0 cm spinal cord tissue was cut for immunohistochemistry and the blood was collected from heart.The expression of MMP-2 was detected in the tissue and serum.The behavior changes of the rabbits were evaluated by the inclined plate test and the modified Tarlov score at different time points.Results:1)The expression of MMP-2 and MMP-12 and behavior evaluated in each group were statistically different between baseline and model established(P<0.05).2)After treatment,the expression of MMP-2 and MMP-12 and behavior evaluated in group A,B,and C were statistically different to group D(P<0.05),and group B were higher than group C(P<0.05).Conclusion:Middle dose of Honghua injection is more effective than the other dose,so this dose can be used as a reference dose for treating of chronic injury of spinal cord,and provide the experimental basis for the clinical treatment of cervical spinal cord disease.
