摘要:
目的:通过观察龟甲胶、鹿角胶含药血清对豚鼠膝骨关节炎软骨细胞增殖及其丝裂原活化蛋白激酶激酶(MKK)表达的影响,阐释龟甲胶、鹿角胶对膝骨关节炎的治疗作用。 方法:将龟甲胶组、鹿角胶组、盐酸氨基葡萄糖组、对照组4组含药血清分别对3月龄豚鼠膝关节软骨细胞进行干预,采用MTT法检测含药血清干预后软骨细胞增殖情况,SABC免疫组化检测含药血清干预后软骨细胞MKK的表达。 结果:5%含药血清干预72 h后,软骨细胞的MTT检测结果为:龟甲胶组OD值为0.648±0.09,鹿角胶组为0.442±0.047,盐酸氨基葡萄糖组为0.353±0.053; 对照组为0.236±0.050,差异有统计学意义(P<0.05)。MKK的SABC免疫组化检测OD值为:龟甲胶组0.330±0.018,鹿角胶组0.274±0.020,盐酸氨基葡萄糖组0.223±0.041,对照组0.171±0.041,差异有统计学意义(P<0.05)。结论:1)豚鼠骨关节炎软骨细胞增殖与MKK的表达一致增长; 2)龟甲胶、鹿角胶能有效上调关节软骨细胞MKK的表达,促进软骨细胞增殖,从而修复受损软骨,延缓骨关节炎进程。
Abstract:
Objective: To investigate the effect of tortoise shell glue and deerhorn glue containing serum on the treatment of knee osteoarthritis through detecting its effect on the proliferation of osteoarthritis chondrocytes of guinea pigs' knee and the expressiong of mitogen-activated protein kinase kinase(MKK). Methods: The osteoarthritis chondrocytes of three-months-old guinea pig was interventioned respectively by 4 kinds serum containing drugs(tortoise shell glue group, deerhorn glue group, glucosamine hydrochloride group, and the control group). The proliferation of chondrocytes was detected using MTT method and the expression of MKK of chondrocytes was tested by SABC immunohistochemical method after intervention. Results: The test results of MTT were 0.648±0.09, 0.442±0.047, 0.353±0.053, and 0.236±0.050 in tortoise shell glue group, deerhorn glue group, glucosamine hydrochloride group, and control group respectively at 72 h after 5% serum containing drugs interventioned, the difference was statistically significant(P<0.05). The OD values of MKK were 0.330±0.018, 0.274±0.020, 0.223±0.041, and 0.171±0.041 in tortoise shell glue group, deerhorn glue group, glucosamine hydrochloride group, and control group respectively, the difference was statistically significant(P<0.05). Conclusion: 1)The proliferation of osteoarthritis chondrocytes of guinea pig was positively correlated with the expression of MKK; 2)Tortoise shell glue and deerhorn glue can effectively increase the expression of MKK and promote proliferation of chondrocytes, lead to repair the damaged cartilage and delay the process of osteoarthritis.
