摘要:
目的:探讨益气活血方促进破裂型腰椎间盘突出后重吸收的机制。方法:30只SD大鼠分成模型组Ⅰ、模型组Ⅱ和中药组,模型组予生理盐水灌胃,中药组予益气活血方灌胃,分别于1周、4周后取材,对埋置的椎间盘质量进行称重,采用免疫组织化学法检测Fas和Caspase-3蛋白的表达,Tunel法检测椎间盘凋亡细胞阳性率。结果:中药组椎间盘质量减少较模型组明显(P<0.05),Fas、Caspase-3的表达明显高于模型组Ⅰ、Ⅱ(P<0.05),椎间盘凋亡细胞阳性率也要高于模型组(P<0.05)。结论:益气活血方可以通过Fas通路促进介导髓核细胞凋亡,从而促进重吸收的发生。
Abstract:
Objective:To investigate the mechanism of Yiqi Huoxue decoction on promoting spontaneous resorption of ruptured lumbar disc herniation. Methods:30 Sprague-Dawley rats were divided into model group Ⅰ, model group Ⅱ, and traditional Chinese medicine(TCM)group. The rats were intragastric administration with normal saline in the model group, and with Yiqi Huoxue decoction in the TCM group. The intervertebral disc were drawn and weighed 1 week and 4 weeks respectively after treating. The expression of Fas and Caspase-3 proteins were detected by immune histochemical method, and the rate of apoptotic cells were measured with the Tunel assay method. Results:In TCM group, the quality of intervertebral disc were reduced more than the model group(P<0.05), the expression of Fas and Caspase-3 were significantly higher than the model group(P<0.05), and the rate of apoptotic cells of intervertebral disc was higher than the model group(P<0.05). Conclusion:Yiqi Huoxue decoction could activate the spontaneous resorption of ruptured lumbar disc herniation though the Fas pathway to promote the apoptosis of nucleus pulposus cells.
