摘要:
目的:采用密骨方干预去卵巢骨质疏松骨折Wistar大鼠,观察密骨方对大鼠骨折断端臂板蛋白3A表达的影响,探讨中药密骨方影响骨折愈合的机制。方法:45只3个月龄雌性Wistar大鼠随机分为3组:假手术骨折对照组(Sham,n=15),骨质疏松骨折模型组(OVX,n=15),骨质疏松骨折密骨方组(OVX-M,n=15)。适应性饲养1周后建立骨质疏松模型,OVX组与OVX-M组依次摘除双侧卵巢,Sham组切除卵巢周围同质量的脂肪组织。3组大鼠去卵巢术后5个月,建立骨折模型,手术刀加外力致股骨中部骨折,并复位固定。骨折术后第2天,OVX-M组进行中药密骨方灌胃干预,采用1 mL/100 g的剂量比例进行灌服,1次/d。分别在密骨方干预后第2,4,6周采集骨组织样本,用Western Blot实验方法检测骨折断端臂板蛋白3A含量。结果:Sham组在第2,4,6周呈上升趋势; OVX组在第2,4,6周呈先上升后下降趋势; OVX-M组在第2,4,6周呈先上升后下降趋势。第2周,Sham组与OVX组比较,差异无统计学意义(P=0.223); OVX-M组与 OVX组比较,差异无统计学意义(P=0.066)。第4周,Sham组与OVX组比较,差异无统计学意义(P=0.159); OVX-M组与 OVX组比较,差异无统计学意义(P=0.093)。第6周,Sham组与OVX组比较,差异有统计学意义(P=0.004); OVX-M组与 OVX组比较,差异有统计学意义(P=0.023)。结论:中药密骨方干预可提高大鼠骨折断端臂板蛋白3A的表达,促进骨质疏松骨折愈合。
Abstract:
To observe the expression of protein 3A by the Migufang in the Wistar rats with ovariectomized osteoporosis fracture,and to explore the mechanism of Migufang on fracture healing.Methods:45 female Wistar rats with 3-month-old were randomly divided into 3 groups,the Sham operation fracture control group(Sham,n=15),the osteoporosis fracture model group(OVX,n=15),and the osteoporosis fracture Migufang group(OVX-M,n=15).The osteoporosis model was established after 1 week of adaptive feeding.Bilateral ovaries were removed in the OVX group and the OVX-M group,and adipose tissue of the same weight around the ovaries was removed in the Sham group.5 months after ovariectomy,the central femur fracture model was established by external force and scalpel,and the fracture was reduced and fixed.On the second day after fracture,the Migufang gavage intervention was carried out with ratio of 1 mL/ 100 g and once a day.Bone tissue samples were collected at 2nd,4th and 6th week after the prognosis of Migufang,and the Semaphorin 3A protein was detected and compared by Western Blot.Results:Protein 3A in the Sham group revealed an upward trend from 2nd,4th to 6th week.It in OVX and OVX-M increased first and then decreased.There was no significant difference among these 3 groups(P>0.05)at the 2nd and 4th week.At the 6th week,there was a significant difference between the Sham group vs OVX group(P<0.01)and the OVX-M group vs the OVX group(P<0.05).Conclusion:Migufang intervention may promote the healing of osteoporotic fracture by improve the expression of Semaphorin 3A protein at the broken end of fracture in rats.
