摘要:
目的:观察龙鳖血清对不同来源干细胞生物特性的影响。方法:从滑液和骨髓中分离培养间充质干细胞(MSCs),以龙鳖血清干预MSCs,观察不同来源MSCs的标记物和炎症因子的表达情况,以及向软骨细胞分化能力的影响。结果:不同来源MSCs培养后的细胞形态及标记物表达无差别,原代SF-MSC表达IL-1β、TNF-α和TGF-β1明显高于原代、传代BM-MSC及传代SF-MSC(P<0.01),龙鳖血清可明显抑制SF-MSC分泌炎症因子(P<0.01),而对BM-MSC分泌炎症因子无明显影响;原代SF-MSC表达的胶原蛋白1、2和3均低于原代和传代的BM-MSC(P<0.05),龙鳖血清可明显诱导原代SF-MSC和BM-MSC向成软骨细胞表达(P<0.05)。结论:龙鳖血清对SF-MSC和BM-MSC均有抑制细胞因子释放,诱导SF-MSC和BM-MSC向软骨分化的能力,这对中药在防治骨关节炎机理研究中具有重要的指导意义。
Abstract:
Objective:To observe the effect of serum of dragon turtle on biological characteristics of different sources of mesenchymal stem cells(MSCs).Methods:The MSCs was isolated from the synovial fluid and bone marrow,which were intervention cultured with serum of dragon turtle.The expression of markers and cytokines of MSCs from different sources,as well as the ability of chondrocyte differentiation was detected.Results:The cell morphology and markers of different sources of cultured MSCs was not different.The expression of IL-1β,TNF-α and TGF-β1 in the primary SF-MSC was significantly higher than that of primary,passage of BM-MSC,and passage of SF-MSC(P<0.01).The serum of dragon turtle can inhibit significantly the secretion of inflammatory cytokines in SF-MSC(P<0.01),but not in the BM-MSC.The expression of collagen 1,2 and 3 in were lower primary SF-MSC was lower than that of primary and passage BM-MSC(P<0.05).The serum of dragon turtle can induce primary SF-MSC and BM-MSC generated into chondrogenic(P<0.05).Conclusion:The serum of dragon turtle is effective to inhibit the release of cytokines of SF-MSC and BM-MSC,and is effective to induce SF-MSC and BM-MSC genenrated into chondrogenic.It is importantly to guide prevention and treatment of osteoarthritis.
