摘要:
目的:探讨补肾活血汤及其拆方含药血清调控大鼠骨髓间充质干细胞(BMSCs)增殖活性的作用。方法:体外骨髓贴壁法分离培养大鼠BMSCs,取第3代细胞,免疫荧光法鉴定细胞表面抗原,成骨及成脂诱导证实其多向分化潜能; 实验分为补肾活血汤全方组、补肾组、活血组及空白对照组,运用补肾活血汤及其拆方含药血清干预第3代 BMSCs 48 h,并用四甲基偶氮唑盐比色法(MTT)观察细胞增殖活性,流式细胞仪检测细胞周期。结果:MTT结果显示:体积分数5%及10%补肾活血汤全方含药血清均有促进大鼠BMSCs增殖的作用; 流式细胞仪检测细胞周期显示:运用补肾活血汤含药血清干预后,处于增值分裂期(S+G2+M期)的BMSCs百分率增多,与其他3组比较差异有统计学意义(P<0.05)。结论:补肾活血汤全方含药血清有促进大鼠骨髓间充质干细胞增殖的作用。
Abstract:
Objective: To investigate the effect of drug serum of BuShenHuoXue decoction and its separated prescription medicated proliferation of rat bone marrow mesenchymal stem cells(BMSCs). Methods: Rat BMSCs were cultured by adherence method in vitro. The third generation of cells was collected. Cell surface antigen was identified by immunofluorescence assay. Osteogenesis and adipogenesis were induced to confirm its multi-directional differentiation potential. The study includes BuShenHuoXue decoction group, strengthening kidney group, activating circulation group and control group. The third generation of BMSCs was treated with drug serum of BuShenHuoXue decoction and its separated prescription by 48 hours. Cell proliferation was determined by MTT assay. Cell cycle was examined by flow-cytometric analysis. Results: MTT shows 5% and 10% drug serum of BuShenHuoXue decoction have the ability to stimulate bone marrow mesenchymal stem cell proliferation. Flow-cytometric analysis shows percentage of BMSCs in proliferative stage(S+G2+M stages)was increased when treated with drug serum of BuShenHuoXue decoction, and there were statistical differences among the groups. Conclusion: Drug serum of BuShenHuoXue decoction can stimulate rat bone marrow mesenchymal stem cell proliferation.
