摘要:
目的:探讨甜瓜籽提取物对闭合性股骨骨折模型大鼠骨愈合的作用及其机制。方法:60只SPF级SD大鼠随机分为对照组(30例)和甜瓜籽组(30例)。通过用髓内针法建立大鼠闭合性股骨骨折模型,造模后甜瓜籽组大鼠按照1 mL/100 g标准每天灌胃2%甜瓜籽提取物,连续4周,对照组灌胃等量的生理盐水。于造模后第1周、第2周和第4周分别分离大鼠股骨,采用micro-CT检查骨骼愈合情况,并分析各时期大鼠股骨创面骨密度水平; 检测各时期两组大鼠血清中Ⅰ型胶原酶和骨钙素的表达水平; 通过qPCR和Western Blot法检测实验终点两组大鼠骨痂成骨关键分子BMP2和Smad4的mRNA及蛋白表达水平。结果:与对照组相比,甜瓜籽组大鼠在造模后第1周、第2周和第4周骨骼愈合优于对照组,骨密度水平显著高于对照组,差异有统计学意义(P<0.01); 血清中Ⅰ型胶原酶和骨钙素的表达水平在各时期均显著高于对照组,差异有统计学意义(P<0.01); qPCR和Western Blot法检测结果表明甜瓜籽组大鼠成骨分子BMP2和Smad4 mRNA及蛋白表达均显著高于对照组,差异有统计学意义(P<0.01)。结论:甜瓜籽提取物对闭合性股骨骨折模型大鼠骨愈合有促进作用,可以增加血清中Ⅰ型胶原酶和骨钙素的表达水平,其机制可能与成骨信号通路BMP2/Smad4的调控相关。
Abstract:
Objective:To investigate the efficacy and mechanism of melon seed extract on the bone fracture healing in rats with closed femoral fracture.Methods:60 SPF SD rats were randomly divided into control group(30 cases)and melon seed group(30 cases). The rat model of closed femoral fracture was established by intramedullary needle. After modeling, the rats in the melon seed group were gavage with 2% sweet melon seed extract according to the standard of 1 mL/100 g every day for 4 weeks, and the rats in the control group were given by gavage with the same amount of normal saline. The femurs of rats were separated at the 1st, 2nd and 4th weeks after modeling, and the bone healing was analyzed by micro-CT, and the bone mineral density of femoral wound in rats in different periods was analyzed. The expression levels of type Ⅰ collagenase and osteocalcin in serum of the two groups were detected. The mRNA and protein expression levels of BMP2 and Smad4 were detected by qPCR and Western Blot.Results:Compared with the control group, the bone healing of melon seed group was better than control group at the 1st, 2nd and 4th weeks after modeling, and the bone mineral density level was significantly higher than control group(P<0.01). The expression levels of type Ⅰ collagenase and osteocalcin in serum were significantly higher than those in the control group(P<0.01). The results of qPCR and Western Blot showed that the expressions of BMP2 and Smad4 mRNA and protein in melon seed group were significantly higher than control group(P<0.01).Conclusion:Melon seed extract can promote bone healing in rats with closed femoral fracture, and it can increase the expression levels of type Ⅰ collagenase and osteocalcin in serum. It may be related to the regulation of osteogenic signal pathway BMP2/Smad4.
