摘要:
目的:观察益肾养髓方对慢性脊髓压迫大鼠模型受压脊髓组织小胶质细胞极化的影响。方法:50只Sprague-Dawley(SD)大鼠随机分为假手术组、模型组、益肾养髓方高、中、低剂量组,采用吸水膨胀材料压迫脊髓的方法进行造模,给药后分别通过斜板试验、苏木精-伊红(HE)染色、蛋白质印迹(Western Blot)法检测、实时荧光定量逆转录聚合酶链式反应(RT-qPCR)评价大鼠的运动功能与CD86、CD206蛋白及IL-18、IL-1β mRNA的表达。结果:各时间节点的中药灌胃组、模型组斜板试验结果与假手术组相比差异均有统计学意义(P<0.05)。中药中、高剂量组斜板试验结果均优于模型组,差异有统计学意义(P<0.05); HE染色可见假手术组神经元细胞形态基本正常,模型组大量神经元细胞受损,细胞肿胀坏死,给药组神经元空泡样改变减少,细胞形态及数量有一定恢复。Western Blot法检测结果显示:与假手术组比较,模型组、中药各给药组CD86蛋白表达均高于假手术组,差异有统计学意义(P<0.05); 中药中、高剂量组CD206表达高于模型组,差异有统计学意义(P<0.05)。中药中、高剂量组CD86/CD206低于假手术组和模型组,差异均有统计学意义(P<0.05)。RT-qPCR结果显示:中药中、高剂量组IL-18表达低于模型组,差异有统计学意义(P<0.05); 中药中、高剂量组IL-1β表达低于模型组,差异有统计学意义(P<0.05)。结论:益肾养髓方可促使脊髓组织中M1型小胶质细胞向M2型分化,减轻脊髓受压时炎症反应,在一定程度上改善大鼠运动功能。
Abstract:
Objective:To observe the efficacy of Yishen Yangsui formula on the polarization of microglia in compressed spinal cord tissue of rats with chronic spinal cord compression.Methods:50 Sprague-Dawley rats were randomly divided into Sham operation group, model group, high, middle and low dose groups of Yishen Yangsui formula. The model was made by compressing the spinal cord with water swelling material. After administration, the motor function and the expressions of CD86, CD206, IL-18 and IL-1β were evaluated by inclined plate test, hematoxylin-eosin(HE)staining, Western Blot and real-time fluorescence quantitative reverse transcriptase polymerase chain reaction(RT-qPCR).Results:There were significant differences in the results of inclined plate test between traditional Chinese medicine intragastric administration group and model group and Sham operation at each time point(P<0.05). The inclined plate test of middle concentration group and high concentration group were better than that of model group(P< 0.05); HE staining showed that the morphology of neurons in the Sham operation group was basically normal. A large number of neurons in the model group were damaged, swelling and necrosis. The vacuolar changes of neurons in the treatment group decreased, and the morphology and number of cells recovered to a certain extent. The results of Western Blot showed that compared with the Sham operation group, the expression of CD86 protein in the model group and traditional Chinese medicine group was higher than that in the Sham operation group(P<0.05), and the expression of CD206 in the middle dose group and high dose group was higher than that in the model group(P<0.05). The expression of CD86/CD206 in middle dose group and high dose group was lower than that in Sham operation group and model group, and the difference was statistically significant(P<0.05). The results of RT-qPCR showed that the expression of IL-18 in the middle dose group and high dose group was lower than that in the model group(P<0.05), and the expression of IL-1β in the middle dose group and high dose group was lower than that in the model group(P<0.05).Conclusion:The Yishen Yangsui formula can promote the differentiation of M1 microglia into M2 in spinal cord tissue, reduce the inflammatory reaction during spinal cord compression and improve the motor function of rats to some extent.
