摘要:
目的:探讨中药青娥方调节MMP2/EGFR通路影响成骨细胞凋亡的作用机制。方法:成骨细胞分为6组,A组只加入培养液。除A组外,均加入30 ng/mL的TNF-α培养液,C组 再加入MMP2重组蛋白,D组再加入5%含药血清,E组再加入10%含药血清,F组 再加入MMP2重组蛋白干预后5%含药血清。最后测定各组凋亡率和信号蛋白表达水平。结果:B组比 A组细胞凋亡数目增加显著,F组凋亡细胞比例最小(仅为22.97%),而 D组中处于 G0-G1期的细胞最少,S期的细胞最多。 C组中ERK2和Bad表达显著减少,差异有统计学意义(P<0.05)。D和E两组比较,EGFR含量轻微下降,ERK2含量稍有增加,而Bad含量无变化,差异均无统计学意义(P>0.05)。F组中EGFR表达显著降低,差异有统计学意义(P<0.01); ERK2和Bad明显增加,差异有统计学意义(P<0.01)。结论:只有中药青娥丸和基质金属蛋白酶同时应用可降低EGFR-ERK2-Bad通路蛋白表达,成骨细胞失凋亡,促进成骨。
Abstract:
Objective:To observe the mechanism of Qinge prescription regulating the apoptosis of osteoblasts by MMP2/EGFR signaling pathway.Methods:Osteoblasts were divided into 6 groups.Group A only added culture medium.The 30 ng/mL TNF-a medium was added in all group except group A.Then, group C was added MMP2 recombinant protein, group D was added 5% drug-containing serum, group E was added 10% drug-containing serum, and group F was added 5% drug-containing serum after intervention with MMP2 recombinant protein.Finally, the apoptosis rate and signal protein level of each group were measured.Results:The number of apoptotic cells in group B was significantly higher than that in group A.The percentage of apoptotic cells in group F was the least, only 22.97%, while that in group D was the least in G0-G1 stage and the largest in S stage.The expression of ERK2 and Bad decreased significantly in group C(P<0.05).The content of EGFR decreased slightly, the content of ERK2 increased slightly, but the content of Bad remained unchanged(P>0.05).The expression of EGFR in group F was significantly decreased(P<0.01), and ERK2 and Bad increased significantly(P<0.01).Conclusion:In this study, only Qing'e prescription and matrix metalloproteinase are used simultaneously to reduce the expression of EGFR/ERK2/Bad pathway protein, osteoblasts lost apoptosis and promote osteogenesis.
