摘要:
目的:探讨血清和关节滑液中Dickkopf相关蛋白1(DKK-1)水平与膝骨关节炎(KOA)患者影像学分级严重程度的相关性。方法:纳入50例KOA患者为观察组,同期健康体检者20例为对照组,采用ELISA法检测两组血清和观察组关节液中DKK-1水平。膝骨关节炎影像学严重程度分级采用Kellgren-Lawrence评分系统。分析血清和膝关节滑液DKK-1水平与KOA严重程度的相关性。结果:KOA患者不同分级组及正常对照组血清DKK-1水平比较,差异有统计学意义(F=13.85,P<0.001); 不同分级KOA患者血清及关节滑液DKK-1水平差异有统计学意义(F=17.678,P<0.001; F=13.978,P<0.001); 血清及关节滑液DKK-1水平随着K-L分级越高而降低(P<0.05); 血清和关节滑液中DKK-1水平与K-L分级有相关性(r=-0.731,P<0.001; r=-0.690,P<0.001); 血清DKK-1水平与关节滑液DKK-1水平呈正相关性(r=0.725,P<0.001)。结论:KOA患者血清和关节滑液DKK-1水平显著降低,与K-L分级呈负相关。
Abstract:
Abstract Objective:To investigate the correlationship between the level of serum and synovia Dickkopf-1(DKK-1)and radiographic grading of Knee Osteoarthritis(KOA). Methods:50 patients with KOA were enrolled into this study as observation group, and 20 healthy subjects as the control group. The levels of serum and synovia DKK-1 were determined by enzyme-linked immunosorbent assay. Radiographic grading of KOA was performed according to the Kellgren-Lawrence(K-L)classification. Correlations between the level of serum and synovia DKK-1 and severity of KOA were analyzed. Results: The serum DKK-1 levels of KOA patients were significantly different between grading groups and normal control group(F=13.85, P<0.001). The levels of serum and synovia DKK-1 between patients with different grades of KOA were significantly different(F=17.678,P<0.001; F=13.978,P<0.001).The levels of serum and synovia DKK-1 decreased with increasing severity of K-L grading(P<0.05 or 0.01). The levels of serum and synovia DKK-1 were negitively correlated with the severity grade of K-L(r=-0.731, P<0.001; r=-0.690,P<0.001). The levels of serum DKK-1 was positively correlated with the levels of synovia DKK-1(r=0.725, P<0.001). Conclusion: The levels of serum and synovia DKK-1 were extremely decreased in the patients with KOA, which were negitively correlated with K-L classification.
