摘要:
目的:探讨和营通气片对大鼠腰椎间盘突出症(LDH)寒凝证的影响,基于NLRP3/NF-κB炎症小体信号通路探讨其可能的作用机制。方法:将30只SD雄性大鼠进行腰椎间盘突出症造模,然后随机挑选24只进行寒冷刺激,随后分成对照组(LDH大鼠模型)、模型组(LDH寒凝证大鼠模型)、和营通气片低剂量组、和营通气片高剂量组及吲哚美辛组,每组6只,干预28 d。用神经功能评分评价寒邪对腰椎间盘突出症的影响,用ELISA法检测IL-1β和TNF-α的含量,用Western Blot法和实时PCR检测NLRP3、NF-κB、ASC、IL-1β、TNF-α蛋白和mRNA的表达水平,用流式细胞仪检测细胞凋亡情况。结果:寒邪促进腰椎间盘突出症的发展,与对照组比较,模型组髓核组织PLA2活性升高,痛阈降低,神经根组织中NLRP3、NF-κB、ASC、IL-1β、TNF-α 蛋白和mRNA表达水平均提高,细胞凋亡增加(P<0.05); 与模型组比较,吲哚美辛组及和营通气片组大鼠神经根组织中髓核组织PLA2活性降低,痛阈升高,NLRP3、NF-κB、ASC、IL-1β、TNF-α 蛋白和mRNA表达水平均降低,细胞凋亡减少(P<0.05)。结论:和营通气片可能通过抑制NLRP3/NF-κB相关表达,缓解腰椎间盘突出症寒凝证大鼠髓核炎症及细胞凋亡,提高大鼠痛阈,NLRP3/NF-κB信号调控通路是和营通气片对大鼠腰椎间盘突出症寒凝证的可能作用机制。
Abstract:
Objective:To explore the effect of Heyingtongqi tablets on cold coagulation syndrome in rats with lumbar disc herniation(LDH),and exploring its possible mechanism of action based on the NLRP3/NF-κB inflammasome signaling pathway.Methods:30 SD male rats were modeled for lumbar disc herniation,and 24 rats were randomly selected for cold stimulation.Then they were divided into control group(LDH rat model),model group(LDH rat model of cold coagulation syndrome),Heyingtongqi tablet low-dose group,Heyingtongqi tablet high-dose group and indomethacin group,with 6 animals in each group.The intervention lasted for 28 d.Neural function score was used to evaluate the effect of cold evil on LDH,ELISA was used to detect the contents of IL-1β and TNF-α,and Western Blot and real-time PCR were used to detect the expression levels of NLRP3,NF-κB,ASC,IL-1β and TNF-α protein and mRNA.The apoptosis was detected by flow cytometry.Results:Cold pathogens promoted the development of LDH.Compared with the control group,PLA2 activity in nucleus pulposus tissue was increased,pain threshold was decreased,NLRP3,NF-κB,ASC,IL-1β,TNF-α protein and mRNA expression levels in nerve root tissue were increased in the model group,and apoptosis was increased(P<0.05).The activity of PLA2 in nucleus pulposus decreased,the pain threshold increased,the expression levels of NLRP3,NF-κB,ASC,IL-1β,TNF-α protein and mRNA decreased,and the apoptosis decreased(P<0.05)in intomethacin group and Heyingtongqi tablet group.Conclusion:Heyingtongqi tablets may alleviate nucleus pulposus inflammation and cell apoptosis in LDH cold coagulation syndrome rats by inhibiting NLRP3/NF-κB related expression,and increase the pain threshold of rats.Therefore,the NLRP3/NF-κB signaling regulatory pathway is a potential mechanism of Heyingtongqi tablets in treating cold coagulation syndrome of lumbar disc herniation in rats.
