摘要:
目的:初步探讨补肾活血方对激素性股骨头缺血性坏死(INFH)小鼠肾素-血管紧张素系统(RAS)经典轴血管紧张素转换酶(ACE)/血管紧张素Ⅱ(AngⅡ)/血管紧张素Ⅱ-1型受体(AT1R)的影响。方法:将60只SPF 级C57BL/6雄性小鼠随机分5组,即健康对照组、激素性INFH模型组、补肾活血方组、骨化三醇组及阿利吉仑组。治疗3个月后,ELISA法检测循环血中1,25(OH)2D3及AngⅡ的含量,并检测股骨头局部VDR、ACE、AT1R mRNA表达情况,行免疫组化分析。结果:与模型组相比,健康对照组、补肾活血方组、骨化三醇组及阿利吉仑组循环血中1,25(OH)2D3含量明显升高,差异有统计学意义(P<0.05),而AngⅡ含量明显降低,差异有统计学意义(P<0.05)。与模型组相比,健康对照组、补肾活血方组、骨化三醇干预组及阿利吉仑组骨组织局部VDR mRNA的表达明显升高,差异有统计学意义(P<0.05),而ACE、AT1R表达明显降低,差异有统计学意义(P<0.05)。免疫组化结果显示:模型组骨组织中ACE及AT1R阳性表达较其他各组高,差异有统计学意义(P<0.05),而VDR阳性表达较其他各组低,差异有统计学意义(P<0.05)。结论:激素性INFH小鼠RAS经典轴ACE/AngⅡ/AT1R被激活,补肾活血方可激活1,25(OH)2D3/VDR信号通路,并能抑制激活的RAS经典轴ACE/AngⅡ/AT1R,这可能是补肾活血方防治激素性INFH的部分机制。
Abstract:
Objective:To study the efficacy of Bushen Huoxue recipe on ACE/AngⅡ/AT1R of steroid induced ischemic necrosis of femoral head(INFH)in mice.Methods:All 60 SPF C57BL/6 male mice were randomly divided into 5 groups:control group,steroid induced INFH model group,Bushen Huoxue recipe group,calcitriol intervention group and aliskiren intervention group.The levels of 1,25(OH)2D3 and AngⅡ in the circulating blood were detected by ELISA method; the expression of VDR,ACE,AT1R mRNA in the femoral head was detected,and immunohistochemistry was performed three months after treatment.Results:Compared with the model group,the content of 1,25(OH)2D3 in the circulating blood of the healthy control group,Bushen Huoxue recipe group,calcitriol intervention group and aliskiren group was increased significantly(P<0.05),while the content of AngⅡ was decreased significantly(P<0.05).Compared with the model group,the expression of VDR mRNA in bone tissue of healthy control group,Bushen Huoxue recipe group,calcitriol intervention group and aliskiren group was increased significantly(P<0.05),while the expression of ACE and AT1R was decreased significantly(P<0.05).The immunohistochemistry results were showed that the expression of ACE and AT1R in bone tissue of model group was higher than that of other groups(P<0.05),while the expression of VDR was lower than that of other groups(P<0.05).Conclusion:ACE/AngⅡ/AT1R of RAS classic axis in steroid induced INFH mice is activated.Bushen Huoxue recipe can activate 1,25(OH)2D3/VDR signal pathway and inhibit the activated ACE/AngⅡ/AT1R of RAS classic axis,which may be part of the mechanism of Bushen Huoxue recipe in the prevention and treatment of steroid induced INFH.
