摘要:
目的:探讨电针治疗对脊髓损伤大鼠神经细胞自噬、凋亡及运动功能的影响。方法:将36只SD大鼠按照随机数字表法分为假手术组、脊髓损伤组、督脉电针组,每组12只。采用MASCIS Impacto1精确撞击制作脊髓损伤模型。假手术组只行椎板切除术,不造成脊髓损伤; 脊髓损伤组采用MASCIS Impacto1法制作脊髓损伤模型,不进行干预治疗; 督脉电针组在脊髓损伤后给予督脉电针治疗。于损伤后第1,3,7天分别对各组大鼠进行BBB运动功能评分。损伤后第7天取脊髓组织,用Western Blot法检测微管相关蛋白1轻链3(LC3)-Ⅱ的变化,用TUNEL法检测细胞凋亡情况。结果:损伤后第3天和第7天,督脉电针组BBB评分为(5.8±0.9)和(9.5±0.9),与脊髓损伤组(4.1±0.8)和(6.4±0.7)分比较显著升高,差异有统计学意义(P<0.05),而术后第1天两者评分为(2.8±0.7)和(2.4±0.5),差异无统计学意义(P>0.05)。与假手术组相比,脊髓损伤组LC3-Ⅱ表达明显升高,差异有统计学意义(P<0.05),细胞凋亡显著增加,差异有统计学意义(P<0.05); 与脊髓损伤组相比,督脉电针组LC3-Ⅱ显著升高,差异有统计学意义(P<0.05),细胞凋亡显著降低,差异有统计学意义(P<0.05)。结论:督脉电针治疗能增强脊髓损伤大鼠神经细胞自噬,减少凋亡,改善大鼠运动功能。
Abstract:
Objective:To explore the effect of electro acupuncture(EA)onautophagy,apoptosis and locomotor function after spinal cord injury in rats.Methods:Thirty-six adult female SD rats were randomly divided into three groups:Sham surgery group,spinal cord injury(SCI)group and Du meridian EA group,with 12 rats in each group.Spinal cord injury model was established using precision impacting with MASCIS Impactol.The Sham group only received laminectomy.The rats in SCI group were sustained SCI and were given no intervention.The rats in Du meridian EA group received Du meridian EA treatment after SCI.Scores of Basso Beattie Bresnahan(BBB)motor functions were assessed at 1,3,7 d after the injury.Then the rats were sacrificed at 7 d after the injury,the expression of LC3-Ⅱ in spinal cord was detected with Western Blot,and the number of apoptotic cells were assessed with TUNEL staining.Results:Scores of BBB scale at 3,7 d after injury in Du meridian EA group(5.8±0.9,9.5±0.9)were increased compared with those in Sham group(4.1±0.8,6.4±0.7)(P<0.05),but there were no significant difference between two groups(2.8±0.7 vs 2.4±0.5,P>0.05)at 1 d after injury.The expression of LC3-Ⅱ and the number of apoptotic cells in SCI group were increased compared with those in Sham group(P<0.05).The expression of LC3-Ⅱ were increased while the number of apoptotic cells were decreased in Du meridian EA group compared with those in SCI group(P<0.05).Conclusion:Du meridian electro acupuncture may enhance autophagy and inhibit apoptosisto improve the locomotor function after spinal cord injury in rats.
