摘要:
目的:观察桃红四物汤对大鼠尾血管吻合模型Notch3受体及血管内皮细胞(Vascular Endothelial Cell,VEC)增殖的影响。方法:将40只实验大鼠适应性喂养1周后随机分为4组,即桃红四物汤组、生理盐水组、假手术组和空白对照组,每组10只。桃红四物汤组和生理盐水组造模后干预,假手术组切开显露血管后直接缝合,空白对照组不做处理,并于术后第2天和第5天两个时间点随机选取5只大鼠,采用脊椎脱臼法处死,切取血管吻合区血管,利用免疫组化检测Notch3的表达情况,并对大鼠外周血分离、获取单个核细胞,进行体外细胞培养,反复纯化后利用MTT法检测VEC增殖情况。结果:桃红四物汤组血管吻合区Notch3蛋白表达含量明显高于生理盐水组、假手术组和空白对照组,差异有统计学意义(P<0.05); 桃红四物汤组VEC增殖明显高于生理盐水组、假手术组和空白对照组,差异有统计学意义(P<0.05)。结论:桃红四物汤能上调血管吻合区血管新生相关因子Notch3受体的表达,能够促进血管内皮细胞的增殖,从而促进血管的成活。
Abstract:
Objective:To observe the effect of Taohong Siwu decoction on the proliferation of Notch3 receptor and vascular endothelial cell(VEC)in rat vascular anastomosis model.Methods:After 1 week of adaptive feeding,40 rats were randomly divided into Taohong Siwu decoction group,normal saline group,sham group and blank control group,10 rats in each group.Taohong Siwu decoction group and normal saline group were intervened after modeling.The sham operation group were directly sutured after incision and exposure of vascular,and blank control group received no treatment.At the 2nd and 5th day after the operation,5 rats were randomly selected from each group and killed by spinal dislocation.The expression of Notch3 was detected by immunohistochemical staining after the anastomosis vessel was cut.After isolation of the peripheral blood of rats,mononuclear cells were obtained and cultured in vitro.After repeated purification,the proliferation of VEC was detected by MTT method. Results:The expression of Notch3 protein of Taohong Siwu decoction group was significantly higher than that of saline group,sham operation group and blank control group(P<0.05),and the difference was statistically significant.The proliferation of VEC of Taohong Siwu decoction group was significantly higher than that of saline group,sham operation group and blank control group(P<0.05),and the difference was statistically significant.Conclusion:Taohong Siwu decoction can increase the expression of angiogenesis related factor of Notch3 receptors in vascular anastomosis area,and promote the proliferation of vascular endothelial cells,thereby promoting the survival of blood vessels.
