摘要:
S信号转导通路的影响,以探讨加味青娥丸治疗激素性股骨头坏死的机制。方法:8周龄SPF 级C57BL/6雄性小鼠随机分5组,每组12只,即空白对照组(A组)、激素性骨坏死模型组(B组)、青娥丸加味组(C组)、骨化三醇干预组(D组)及血管紧张素转换酶抑制剂(ACEI)干预组(E组),治疗半年后,运用micro-CT检测并经三维重建获得股骨头颈局部骨组织微观结构,检测股骨头颈局部1,25(OH)2D3/VDR mRNA/RAS系统(ACE,AngⅡ mRNA)表达情况。结果:空白对照组(A组)骨体积分数(BV/TV)、骨小梁数目(Tb.N)、骨小梁厚度(Tb.Th)较模型组(B组)显著增加,差异有统计学意义(P<0.05),结构模型指数(SMI)、骨小梁间隙(Tb.Sp)、骨表面积体积比(BS/BV)较模型组(B组)明显降低,差异有统计学意义(P<0.05),模型组股骨头局部骨小梁稀疏,孔隙率增高,结构紊乱,部分骨小梁结构不完整,表明模型成功。治疗后C组、D组及E组1,25(OH)2D3,VDR mRNA表达均较模型组高,差异有统计学意义(P<0.05),而ACE及AngⅡ mRNA表达较模型组低,差异有统计学意义(P<0.05)。结论:加味青娥丸可能通过调节1,25(OH)2D3/VDR mRNA/RAS信号转导通路而对股骨头坏死疾病的进展起到一定的治疗作用。
Abstract:
Objective: To observe the effects of modified Qing'e Pill on the local signal transduction pathway of 1,25(OH)2D3/VDR mRNA/RAS in mice of steroid-induced ischemic necrosis of femoral head, and to explore the mechanism of modified Qing'e pill in the treatment of steroid induced necrosis of femoral head.Methods: All 60 SPF male C57BL/6 mice, 8-weeks-old, were randomly divided into 5 groups, 12 rats in each group: control group(group A), steroid induced osteonecrosis model group(group B), Qing'e pill group(group C), calcitriol intervention group(group D)and inhibitor of angiotensin converting enzyme(ACEI)intervention group(group E).Six months later, micro-CT detection and three-dimensional reconstruction were used to obtain the local bone tissue of femoral head, and the level of 1,25(OH)2D3, VDR mRNA and RAS system(ACE and AngⅡ mRNA)in the femoral head were detected.Results: The bone volume fraction(BV/TV), the number of trabeculae(Tb.N)and the thickness of the trabecular bone(Tb.Th)in the blank control group A increased significantly compared with the model group(group B)(P<0.05), but the structure model index(SMI), trabecular space(Tb.Sp)and bone surface area volume ratio(BS/BV)were significantly lower(P<0.05).In the model group, the local trabecular bone of the femoral head was sparse, the porosity was increased, the structure was disturbed and some trabecular structures were incomplete.The expressions of 1,25(OH)2D3 and VDR mRNA in group C, group D and group E were higher than those in model group(P<0.05).The expression of ACE and AngⅡ mRNA in group C, D and E was lower than that in model group(P<0.05).Conclusion: Modified Qing'e pill is effective for treating the steroid-induced ischemic necrosis of femoral head through the local signal transduction pathway of 1,25(OH)2D3/VDR mRNA/RAS.
