摘要:
目的:通过实验观察龟甲胶、鹿角胶含药血清对豚鼠膝骨关节炎软骨细胞促增殖作用以及两种含药血清对JNK及p38 MAPK基因表达的影响,揭示龟甲胶及鹿角胶治疗膝骨关节炎的作用机制。方法:分别用龟甲胶、鹿角胶、盐酸氨基葡萄糖(GHC)含药血清及生理盐水对3月龄豚鼠骨关节炎软骨细胞进行干预,MTT法比较含药血清对软骨细胞的促增殖情况,QPCR检测含药血清干预后软骨细胞JNK及p38 MAPK基因的表达情况。结果:用含药血清浓度为5%的各组培养液干预后(时间72 h),龟甲胶组、鹿角胶组、氨基葡萄糖组和空白对照组MTT法检测OD值分别为0.468±0.017,0.425±0.010,0.337±0.017,0.276±0.027,结果显示差异有统计学意义(P<0.05)。QPCR检测龟甲胶组、鹿角胶组、盐酸氨基葡萄糖组和空白对照组JNK mRNA相对表达量分别为0.162±0.096,0.333±0.165,0.653±0.141,1.000±0.000,差异有统计学意义(P<0.05)。QPCR检测龟甲胶组、鹿角胶组、盐酸氨基葡萄糖组和空白对照组p38 MAPK mRNA相对表达量分别为0.058±0.025,0.229±0.057,0.441±0.090,1.000±0.000,结果显示差异有统计学意义(P<0.05)。结论:龟甲胶和鹿角胶都能有效减少豚鼠骨关节炎软骨细胞JNK及p38 MAPK基因的表达,对软骨细胞作用为促增殖,对其凋亡产生抑制作用,从而一定程度上减缓骨关节炎的病损进展。
Abstract:
Objective:To observe serum containing tortoise shell glue and deer-horn glue on the proliferation of chondrocytes and expression of JNK and p38 MAPK gene in osteoarthritis chondrocytes of guinea pigs, and to preliminarily reveal the mechanism in the treatment of knee osteoarthritis. Methods:The osteoarthritis chondrocytes of three months' guinea pigs were cultured intervened with four different serum containing drugs group, such as tortoise-shell glue group, deer-horn glue group, glucosamine hydrochloride group, and blank control group. The effect of chondrocytes proliferation was examined by MTT method, and the expression of JNK and p38 MAPK gene in osteoarthritis chondrocytes of four groups was detected by QPCR method. Results:The OD value determined by MTT method was 0.468±0.017, 0.425±0.010, 0.337±0.017, and 0.276±0.027 in the tortoise-shell glue group, deer-horn glue group, glucosamine hydrochloride group, and blank control group respectively 72 hours after intervened with 5% serum containing drugs. There was significantly difference(P<0.05). The relative expression of JNK mRNA was 0.162±0.096, 0.333±0.165, 0.653±0.141, and 1.000±0.000 respectively in the four groups, and the relative expression of p38 MAPK mRNA was 0.058±0.025, 0.229±0.057, 0.441±0.090, and 1.000±0.000 respectively in the four groups. There was significantly difference(P<0.05). Conclusion:Tortoise-shell glue and deer-horn glue can effectively reduce the expression of JNK and p38 MAPK mRNA in osteoarthritis chondrocytes of guinea pigs, and can inhibit apoptosis and promote proliferation of cartilage cell, leading to slow down the process of osteoarthritis.
