摘要:
目的:探讨补阳还五汤对脊髓损伤后轴突再髓鞘化与BMPR(骨成型蛋白受体)Ⅰa,Ⅱ蛋白表达的影响。方法:将90只大鼠随机分为补阳还五汤组、模型组、假手术组,每组再分为3小组,每小组10只; 假手术组单纯咬除T10椎板,不损伤脊髓,模型组和补阳还五汤组采用自制改良的Allen's撞击器来制备T10节段脊髓损伤大鼠模型,术后补阳还五汤组给予补阳还五汤浓缩液灌胃4周,各组大鼠于造模后1 d、1周、2周、3周、4周进行行为学BBB功能评分(Basso,Beattie & Bresnalan locomotor rating scale),于造模后1周、2周、4周,随机选取每组中的每1小组,处死后取T10脊髓组织,通过苏木精-伊红染色观察脊髓组织形态学改变,通过免疫组化检测BMPR Ⅰa,Ⅱ蛋白表达情况。结果:造模后4周,补阳还五汤组大鼠BBB评分明显高于模型组,P<0.05,差异有统计学意义。分析免疫组化检测结果提示:造模后1周、2周、4周,模型组脊髓损伤节段BMPR Ⅰa,Ⅱ蛋白表达量大量增加,假手术组BMPR Ⅰa蛋白少量表达,补阳还五汤组BMPR Ⅰa,Ⅱ蛋白表达少于模型组,各组组间差异有统计学意义(P<0.05)。结论:补阳还五汤可作用于脊髓损伤后BMP(骨形态发生蛋白)信号通道的相关蛋白,并对其有一定的抑制作用,降低BMPR Ⅰa,Ⅱ受体的表达,促进脊髓损伤后残存轴突再髓鞘化,进而促进神经功能修复。
Abstract:
Objective:To investigate the effect of Buyanghuanwu decoction(BYHWT)on expression of BMPR Ⅰa,Ⅱ protein and axonal remyelination after spinal cord injury.Methods:All 90 rats were randomly divided into BYHWT group,model group,and sham operation group(n=30),each group was further divided into 3 sub-groups(n=10).The T10 of rats in sham group were resection without spinal cord injury,and the rats in model group and BYHWT group were established T10-segment model of spinal cord injury model by modified Allen's combat method.The rats in BYHWT group were then received BYHWT intragastric gavage for 4 weeks.The BBB score were recorded at 1 d,1 week,2 weeks,3 weeks,and 4 weeks after modeling.The T10 spinal cord was executed from each sub-group at 1 week,2 weeks,and 4 weeks after modeling,then the change of tissue morphology was detected by H-E staining,and the expression of BMPR Ⅰa,Ⅱ protein were test by immunohistochemistry.Results:The BBB score in BYHWT group was significantly higher than the model group 4 weeks after modeling(P<0.05).The BMPR Ⅰa,Ⅱ protein in spinal cord injury segment was significantly increased expression 1 week,2 weeks,and 4 weeks after modeling,and was less expression in sham operation group.The expression of BMPR Ⅰa,Ⅱ protein in BYHWT group was lower than model group.There was significant difference between each two groups(P<0.05).Conclusion:BYHWT can be applied in spinal cord injury though BMP signaling pathway.It can reduce the expression of BMPR Ⅰa,Ⅱ receptor,promote remyelination of the surviving axons after spinal cord injury,and thus promote the repair of nerve function.
