摘要:
目的:探讨牛膝有效成分牛膝竹节参皂苷与杜仲有效成分杜仲松脂醇二葡萄糖苷联合应用对骨质疏松性骨折的疗效及作用机理。方法:6月龄成年雌性Wistar大鼠随机分为3组:假手术对照组,模型组,治疗组。治疗组采用灌胃方式给予牛膝竹节参皂苷与杜仲松脂醇二葡萄糖苷混合溶液100 mg/(kg·d), 对照组和模型组给予同量的生理盐水。分别于2、4、6周处死模型鼠,取骨折端组织以及血清检测相关指标。结果:牛膝竹节参皂苷与杜仲松脂醇二葡萄糖苷联合应用治疗能够有效降低去卵巢Wistar大鼠骨质疏松性骨折模型血清中骨钙素(Bone Gla Protein, BGP)和碱性磷酸酶(Alkalin phosphatase, ALP)含量,显著上调骨折端护骨素(Osteoprotegerin, OPG)基因表达水平,但是明显下调骨形态发生蛋白-3(BMP-3)基因表达。结论:牛膝竹节参皂苷与杜仲松脂醇二葡萄糖苷联合应用能有效治疗雌激素缺乏所导致的大鼠骨质疏松性骨折。
Abstract:
Objective: To investigate the therapeutic effect and the mechanism of combined chikusetsu saponin of achyranthes bidentata blume and pinoresinol diglucoside of eucommia for treating the osteoporotic fracture(OPF). Methods: The Wistar rats of 6 months were divided into control group, model group and treatment group. The rats of treatment group were administered the mixture of chikusetsu saponin of achyranthes bidentata blume and pinoresinol diglucoside of eucommia(100mg/kg·d)by gavage. The rats of control group and model group were administered the same does of normal saline.The 2nd, 4th, and 6th weeks after treatment, the rats were sacrificed and the blood and the tissues at the fracture site were collected to detect the relevant index. Results: Combined using of chikusetsu saponin of achyranthes bidentata blume and pinoresinol diglucoside of eucommia could decreased the circulation BGP and ALP of ovariectomized osteoporosis rat, could significantly enhanced the expression of OPG mRNA in the tissues of fracture site, and could obviously down-regulated the expression of BMP-3 mRNA. Conclusion: Combined using of chikusetsu saponin of achyranthes bidentata blume and pinoresinol diglucoside of eucommia is effective for treating the OPF caused by estrogen deficiency.
