摘要:
目的:探究积雪草苷脂质体凝胶通过Toll样受体4(TLR4)/核因子κB(NF-κB)通路对糖尿病足溃疡(DFU)大鼠氧化应激的影响。方法:将大鼠随机分为对照组、模型组(复制糖尿病后钕铁硼磁铁按压足背)、药物低剂量组、药物高剂量组及药物高剂量+激活剂组,每组12只。末次给药24 h后,检测创面愈合率; 酶联免疫吸附测定(ELISA)检测血清血管内皮生长因子(VEGF)、肿瘤坏死因子-α(TNF-α),以及创面组织碱性成纤维细胞生长因子(bFGF)和血管性血友病因子(vWF)水平; 用试剂盒检测创面组织中超氧化物歧化酶(SOD)及丙二醛(MDA)水平; 苏木精-伊红(HE)染色观察创面组织病理变化; Western Blot法检测创面组织中TLR4/NF-κB通路相关蛋白表达。结果:相较于对照组,模型组创面愈合率、VEGF、bFGF、vWF、SOD水平降低,TNF-α、MDA、TLR4、p-NF-κB/NF-κB水平增高(P<0.05); 相较于模型组,药物低剂量组和药物高剂量组创面愈合率、VEGF、bFGF、vWF、SOD增高,TNF-α、MDA、TLR4、p-NF-κB/NF-κB水平下降(P<0.05); TLR4激活剂可减弱药物高剂量对糖尿病足溃疡大鼠上述指标的影响(P<0.05)。结论:积雪草苷脂质体凝胶可能通过抑制TLR4/NF-κB信号通路,改善糖尿病足溃疡大鼠新生血管生成及氧化应激情况。
Abstract:
Objective:To investigate the impacts of asiaticoside liposome gel on neovascularization and oxidative stress in diabetic foot ulcer(DFU)rats through toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)pathways.Methods:Rats were stochastically assigned into control group,model group(after replicating diabetes,NdFeB magnet pressing on the dorsum of the foot),drug low-dose group,drug high-dose group,and drug high-dose + activator group,with 12 rats in each group.24 h after the last administration,the wound healing rate was measured.Enzyme-linked immunosorbent assay(ELISA)was used to detect vascular endothelial growth factor(VEGF),tumor necrosis faction-α(TNF-α)in the serum,and basic fibroblast growth factor(bFGF)and von Willefibrilia factor(vWF)in wound tissue.The reagent kit was used to detect superoxide dismutase(SOD)and malondialdehyde(MDA)in the wound tissues.The pathological changes of wound tissue was observed by hematoxylin-eosin(HE)staining; Western Blot was used to detect TLR4/NF-κB pathway related protein expression in wound tissue.Results:Compared with the control group,the wound healing rate,VEGF,bFGF,vWF,and SOD levels in model group were lower,while TNF-α,MDA,TLR4,and p-NF-κB/NF-κB were higher(P<0.05).Compared with the model group,the wound healing rate,VEGF,bFGF,vWF,and SOD in drug low-dose group and drug high-dose group were higher,while TNF-α,MDA,TLR4,and p-NF-κB/NF-κB were lower(P<0.05).TLR4 activator can reduce the effects of drug high-dose on DFU rats(P<0.05).Conclusion:Asiaticoside liposome gel may improve neovascularization and oxidative stress in DFU rats by inhibiting TLR4/NF-κB signaling pathway.
