摘要:
目的:探究积雪草苷(AC)对糖尿病足溃疡(DFU)大鼠炎症反应、创面愈合及沉默信息调节因子1(SIRT1)/核因子E2相关因子2(Nrf2)/血红素氧合酶1(HO-1)通路的影响。方法:利用高糖高脂食料喂养,腹腔注射链脲佐菌素(STZ)并联合钕铁硼磁铁按压建立糖尿病足溃疡大鼠模型,随机将大鼠分为糖尿病足溃疡组、积雪草苷低剂量组、积雪草苷高剂量组、积雪草苷高剂量+抑制剂组、二甲双胍组,每组6只,6只正常饲养的大鼠为对照组。测算各组大鼠创面愈合率; 用血糖仪检测各组大鼠敷药后的空腹血糖(FBG)水平; ELISA法检测各组大鼠血清中相关炎症因子水平; 苏木精-伊红(HE)染色观察各组大鼠创面愈合组织的病理变化; qRT-PCR法检测各组大鼠创面愈合组织中SIRT1、Nrf2和HO-1 mRNA表达水平; Western Blot法检测各组大鼠创面愈合组织中SIRT1/Nrf2/HO-1通路相关蛋白表达。结果:糖尿病足溃疡组大鼠的创面愈合率、愈合组织中SIRT1、Nrf2和HO-1 mRNA及其蛋白表达水平显著低于对照组,FBG、IL-1β、IL-6、TNF-α水平显著高于对照组,差异有统计学意义(P<0.05); 二甲双胍组、积雪草苷低剂量组和高剂量组大鼠的创面愈合率、愈合组织中SIRT1、Nrf2和HO-1 mRNA及其蛋白表达水平显著高于糖尿病足溃疡组,FBG、IL-1β、IL-6、TNF-α水平显著低于糖尿病足溃疡组,差异有统计学意义(P<0.05),积雪草苷高剂量组效果优于积雪草苷低剂量组,差异有统计学意义(P<0.05),而积雪草苷高剂量组与二甲双胍组差异无统计学意义(P>0.05)。SIRT1抑制剂回补实验发现,积雪草苷对糖尿病足溃疡大鼠的炎症反应和创面愈合以及SIRT1/Nrf2/HO-1通路作用被逆转(P<0.05)。结论:积雪草苷可能通过激活SIRT1/Nrf2/HO-1信号通路,使糖尿病足溃疡大鼠炎症反应和创面愈合情况得到改善。
Abstract:
Objective:To investigate the efficacy of asiaticoside(AC)on inflammatory response,wound healing and silent information regulator 1(SIRT1)/nuclear factor erythroid 2-related factor 2(Nrf2)/heme oxygenase-1(HO-1)pathway in rats with diabetic foot ulcer(DFU).Methods:The DFU rat model was established by feeding with high-sugar and high-fat diet,intraperitoneal injection of streptozotocin(STZ)and pressing with NdFe-B magnet.The rats were randomly divided into diabetic foot ulcer group,asiaticoside low-dose group,asiaticoside high-dose group,asiaticoside high-dose+inhibitor group,and metformin group,with 6 rats in each group,and the remaining 6 normal rats were used as control group.The wound healing rate of rats in each group was measured.Blood glucose meter was used to detect the fasting blood glucose(FBG)level of rats after dressing.ELISA was used to detect the levels of related inflammatory factors in the serum of rats in each group.HE staining was used to observe the pathological changes of wound healing tissue of rats in each group.qRT-PCR was used to detect the mRNA expression levels of SIRT1,Nrf2,and HO-1 in wound healing tissue of rats in each group.Western Blot was used to detect the expressions of proteins related to SIRT1/Nrf2/HO-1 pathway in wound healing tissue of rats in each group.Results:The wound healing rate and the mRNA and protein expression levels of SIRT1,Nrf2,and HO-1 in healed tissue of rats in the diabetic foot ulcer group were significantly lower than those in the control group,and the levels of FBG,IL-1β,IL-6,and TNF-α were significantly higher than those in the control group(P<0.05).The wound healing rate of rats in metformin group,asiaticoside low-dose group,and asiaticoside high-dose group were significantly higher than those in diabetic foot ulcer group,and the mRNA and protein expression levels of SIRT1,Nrf2,and HO-1 in healed tissue were significantly higher than those in diabetic foot ulcer group,and the levels of FBG,IL-1β,IL-6,and TNF-α were significantly lower than those in diabetic foot ulcer group(P<0.05).The effect of asiaticoside high-dose group was better than that of asiaticoside low-dose group(P<0.05),while there was no significant difference between asiaticoside high-dose group and metformin group(P>0.05).SIRT1 inhibitor complementation experiment showed that the inflammatory response and wound healing as well as the effect of SIRT1/Nrf2/HO-1 pathway on DFU rats were reversed by AC(P<0.05).Conclusion:AC may improve the inflammatory response and wound healing in DFU rats by activating SIRT1/Nrf2/HO-1 signaling pathway.
