摘要:
目的:探讨花青素对腰椎间盘突出症(LDH)大鼠的抗炎镇痛及磷脂酰肌醇3激酶(PI3K)/丝氨酸-苏氨酸激酶(Akt)/核因子-κB(NF-κB)信号通路的作用。方法:将所有大鼠分为对照组、模型组、花青素低/高剂量组、花青素高剂量+PI3K抑制剂组(花青素高剂量+LY294002组),除对照组外,其余所有大鼠构建腰椎间盘突出症大鼠模型,构建模型成功后分组给药处理,进行大鼠神经功能评分,检测大鼠机械性撤足阈值及热撤足反应期阈值,用ELISA法检测大鼠背根神经节组织炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平及神经活性物质5-羟色胺(5-HT)、P物质(SP)水平,用尼氏(Nissl)染色观察大鼠背根神经节组织病理学变化,用Western Blot法检测p-PI3K/PI3K、p-Akt/Akt、p-NF-κB p65/NF-κB p65表达。结果:模型组背根神经节组织形态异常,神经纤维排列散乱,细胞肿胀,空泡严重,尼氏小体紊乱,其神经功能评分和TNF-α、IL-1β、5-HT、SP水平及p-NF-κB p65/NF-κB p65表达高于对照组,机械性撤足阈值、热撤足反应期阈值及p-PI3K/PI3K、p-Akt/Akt表达低于对照组,差异有统计学意义(P<0.05); 花青素低/高剂量组背根神经节组织形态较正常,神经纤维排列较整齐,细胞空泡现象改善,其神经功能评分和TNF-α、IL-1β、5-HT、SP水平及p-NF-κB p65/NF-κB p65表达低于模型组,机械性撤足阈值、热撤足反应期阈值及p-PI3K/PI3K、p-Akt/Akt表达高于模型组,其中花青素高剂量组变化最显著,差异有统计学意义(P<0.05); LY294002可部分逆转花青素对腰椎间盘突出症大鼠的抗炎镇痛作用,差异有统计学意义(P<0.05)。结论:花青素可对腰椎间盘突出症大鼠发挥抗炎镇痛作用,其作用机制与激活PI3K/Akt/NF-κB信号通路有关。
Abstract:
Objective:To investigate the anti-inflammatory and analgesic effects of anthocyanidin on lumbar disc herniation(LDH)rats,and its effects on PI3K/Akt/NF-κB signaling pathway.Methods:All rats were separated into control group,model group,low-dose and high-dose anthocyanin groups,and high-dose anthocyanin+PI3K inhibitor group(high-dose anthocyanin+LY294002 group).Except for the control group,LDH rat models were constructed for all other rats,and after successful construction,they were grouped and treated with medication.The neurological function scoring was performed among rats.The mechanical withdrawal threshold and thermal withdrawal reaction period threshold of rats were detected.ELISA was applied to detect the levels of inflammatory factors such as tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β),and the levels of neurotransmitters serotonin(5-HT)and substance P(SP)in dorsal root ganglion tissue.Nissl staining was applied to observe pathological changes in the dorsal root ganglion tissue of rats.Western Blot method was applied to detect the expression of p-PI3K/PI3K,p-Akt/Akt,and p-NF-κB p65/NF-κB p65.Results:The morphology of the dorsal root ganglion tissue in the model group was abnormal,with scattered arrangement of nerve fibers,cell swelling,severe vacuoles,and disorder of Nissl bodies,the neurological function score,TNF-α,IL-1β,5-HT,SP levels,and p-NF-κB p65/NF-κB p65 expression were higher than those in the control group,the mechanical withdrawal threshold,thermal withdrawal reaction period threshold,and p-PI3K/PI3K,p-Akt/Akt expression were lower than those in the control group(P<0.05).The morphology of the dorsal root ganglion tissue in the low and high dose anthocyanidin groups was relatively normal,the arrangement of nerve fibers was more orderly,and the phenomenon of cell vacuoles was improved,the neurological function score,TNF-α,IL-1β,5-HT,SP levels,and p-NF-κB p65/NF-κB p65 expression were lower than those in the model group,the mechanical withdrawal threshold,thermal withdrawal reaction period threshold,and p-PI3K/PI3K,p-Akt/Akt expression were higher than those in the model group,the high-dose anthocyanidin group showed the most obvious changes(P<0.05).LY294002 was able to partially reverse the anti-inflammatory and analgesic effects of anthocyanidin on LDH rats(P<0.05).Conclusion:Anthocyanidin can exert anti-inflammatory and analgesic effects on LDH rats,and their mechanism of action is related to the activation of the PI3K/Akt/NF-κB signaling pathway.
