摘要:
目的:探讨川陈皮素(NOB)对骨质疏松大鼠骨密度及骨代谢的调节能力及Hippo/YAP信号通路的影响。方法:构建骨质疏松大鼠模型并将其随机分为模型组,低、中、高剂量川陈皮素组,高剂量川陈皮素+通路抑制剂(维替泊芬)组,每组12只大鼠; 另取12只健康大鼠作为对照组。对大鼠进行骨密度(BMD)检测,血清骨代谢指标检测,Micro-CT检测股骨组织微结构变化,TRAP染色观察破骨细胞数量,HE染色观察股骨组织病理形态,Western Blot检测Hippo/YAP信号通路有关蛋白表达。结果:与对照组比,模型组大鼠股骨组织病损程度严重,破骨细胞数量增多,骨密度指数、Tb.Th、Tb.N和BV/TV指数以及血清BALP、PINP和CTX-Ⅰ水平显著下降,Tb.Sp指数以及血清中IL-1β水平显著升高,YAP、p-YAP、TAZ蛋白表达水平显著降低,差异有统计学意义(P<0.05); 与模型组比,随着川陈皮素剂量的增加,低、中、高剂量川陈皮素组大鼠股骨组织病损程度明显减轻,破骨细胞数量减少,骨密度指数、Tb.Th、Tb.N和BV/TV指数以及血清BALP、PINP和CTX-Ⅰ水平逐渐升高,Tb.Sp指数以及血清中IL-1β水平逐渐降低,YAP、p-YAP、TAZ蛋白表达水平显著上调,差异有统计学意义(P<0.05); 与高剂量川陈皮素组相比,高剂量川陈皮素+维替泊芬组大鼠股骨组织病损程度严重,破骨细胞数量增多,骨密度指数、Tb.Th、Tb.N和BV/TV指数以及血清BALP、PINP和CTX-Ⅰ水平,YAP、p-YAP、TAZ蛋白表达水平显著降低,Tb.Sp指数以及血清中IL-1β水平显著升高,差异有统计学意义(P<0.05)。结论:川陈皮素可减轻大鼠股骨组织病损程度,调节骨代谢,改善骨质疏松症状,其作用可能与激活Hippo/YAP信号通路有关。
Abstract:
Objective:To investigate the regulatory ability of nobiletin(NOB)on bone density and bone metabolism in osteoporotic rats,and its effect on Hippo/YAP signaling pathway.Methods:A rat model of osteoporosis was constructed and randomly separated into model group,low,medium,and high dose nobiletin groups(NOB-L,NOB-M,NOB-H groups),and high-dose nobiletin+pathway inhibitor group(NOB-H+VTPF group),with 12 rats in each group.Another 12 healthy rats were selected as the control group.Bone density(BMD)testing and serum bone metabolism index testing were performed on rats.Micro-CT was used to detect changes in the microstructure of femoral tissue.TRAP staining was used to observe the number of osteoclasts.HE staining was applied to observe the pathological morphology of femoral tissue.Western Blot was applied to detect protein expression related to the Hippo/YAP signaling pathway.Results:Compared with the control group,the model group had severe femoral tissue damage and an increase in the number of osteoclasts,the BMD index,Tb.Th,Tb.N,BV/TV indexes,and serum BALP,PINP,and CTX-Ⅰ levels unusually decreased,the Tb.Sp index and serum IL-1β level obviously increased,and the YAP,p-YAP,and TAZ protein expression levels unusually decreased(P<0.05).Compared with the model group,with the increase of NOB dose,the degree of femoral tissue damage and the number of osteoclasts in the NOB-L,NOB-M,and NOB-H groups were unusually reduced,the BMD index,Tb.Th,Tb.N,BV/TV indexes,and serum BALP,PINP,and CTX-Ⅰ levels obviously increased,the Tb.Sp index and serum IL-1β level obviously reduced,and the YAP,p-YAP,and TAZ protein expression levels were unusually upregulated(P<0.05).Compared with the NOB-H group,the NOB-H+VTPF group had severe damage to the femoral tissue and an increase in the number of osteoclasts,the BMD index,Tb.Th,Tb.N,BV/TV indexes,serum BALP,PINP,and CTX-Ⅰ levels,and YAP,p-YAP,and TAZ protein expression levels obviously decreased,the Tb.Sp index and serum IL-1β level unusually increased(P<0.05).Conclusion:Nobiletin can alleviate the degree of femoral tissue damage,regulate bone metabolism,and improve osteoporosis symptoms in rats.Its effect may be related to the activation of the Hippo/YAP signaling pathway.
