摘要:
目的:观察解毒化瘀外洗方联合抗生素对钛表面表皮葡萄球菌(SE)生物膜形成及成熟生物膜的抑制作用。方法:以耐药表皮葡萄球菌ATCC35984为研究对象,采用微量稀释法分别检测解毒化瘀外洗方、左氧氟沙星、头孢替安、克林霉素、庆大霉素及万古霉素的最低抑菌浓度(MIC)以及联合用药部分抑菌浓度指数(FICI),筛选出对表皮葡萄球菌生物膜具有较强抑制作用的药物组合。通过倍比稀释活菌涂板计数、结晶紫染色、扫描电子显微镜(SEM)以及激光共聚焦显微镜(CLSM)观察联合用药对钛表面表皮葡萄球菌生物膜形成及成熟生物膜的影响。结果:解毒化瘀外洗方联合左氧氟沙星是对表皮葡萄球菌生物膜具有较强抑制作用的药物组合; 无论是定性分析还是定量分析,解毒化瘀外洗方联合左氧氟沙星可以抑制钛表面表皮葡萄球菌生物膜的形成和破坏表皮葡萄球菌成熟生物膜。结论:解毒化瘀外洗方联合左氧氟沙星对钛表面表皮葡萄球菌生物膜形成有抑制作用,且早期抑制作用最强; 两药联用可以破坏表皮葡萄球菌成熟生物膜,并且在一定程度上杀灭膜内细菌。
Abstract:
Objective:To observe the inhibitory effect of Jiedu Huayu decoction combined with antibiotics on biofilm formation and mature biofilm of staphylococcus epidermidis(SE)on titanium surface.Methods:Using drug-resistant staphylococcus epidermidis ATCC35984 as the study object,the minimum inhibitory concentration(MIC)and the fractional inhibitory concentration index(FICI)of the combination of Jiedu Huayu decoction,levofloxacin,cefotiam,clindamycin,gentamycin,and vancomycin were detected by using the microdilution method,respectively.Fractional inhibitory concentration index was used to screen out the drug combinations with strong inhibitory effects on SE biofilm.The effects of the combination of drugs on the formation and maturation of SE biofilm on the titanium surface were observed by doubled dilution plate counting,crystal violet staining,scanning electron microscope(SEM)and confocal laser scanning microscope(CLSM).Results:The combination of Jiedu Huayu decoction with levofloxacin was a drug combination with strong inhibitory effect on SE biofilm.Secondly,both qualitative and quantitative analyses showed that the combination of Jiedu Huayu decoction and levofloxacin could inhibit the formation of SE biofilm on the titanium surface as well as destroy the SE mature biofilm.Conclusion:The combination of Jiedu Huayu decoction and levofloxacin can inhibit the formation of SE biofilm on titanium surface,and the strongest inhibitory effect was found in the early stage.Secondly,the combination of the two drugs can destroy SE mature biofilm,and to some extent,it can also kill the bacteria in the membrane.
