摘要:
目的:观察三七总皂苷(PNS)干预股骨头坏死(ONFH)大鼠模型后,PNS对ONFH大鼠股骨头组织成骨作用的影响。方法:36只 SPF 级雄性6个月龄SD大鼠按照随机数字表法分为对照组(Ctrl 组)、模型组(M组)和干预组(PNS组),每组大鼠12只; M组和PNS组行液氮冷冻法股骨头坏死造模,造模3个月后,M组大鼠每日腹腔注射生理盐水2 mL; PNS组大鼠腹腔注射12.0 mg/mL浓度PNS生理盐水溶液2 mL,1次/日,连续两周; Ctrl组不干预。干预后8周各组大鼠处死,采集血清、股骨头标本,行苏木精-伊红(HE)染色、OPG(Osteoprotegerin)及RANKL(Receptor Activator of NF-kB Ligand)Western Blot蛋白质电泳,ELISA法测定PINP(Procollagen Type-I N Propeptide)及TRAP-5b(Tartrate Resistant Acid Phosphatase-5b)血清浓度。结果:PNS干预液氮法ONFH大鼠可降低股骨头空骨陷窝率(P<0.01),可下调骨组织RANKL(P<0.01)及血清TRAP-5b(P<0.05)表达,上调骨组织OPG(P<0.01)表达,可提高血清PINP水平,但差异无统计学意义。结论:PNS可抑制破骨细胞活性,促进成骨,在骨坏死中发挥保护作用。
Abstract:
Objective:To explore the effect of Panax Notoginseng Saponins(PNS)on the osteogenesis of femoral head in rats with osteonecrosis of the femoral head(ONFH).Methods:36 SPF 6-month-old male SD rats were randomly divided into three groups:control group(group Ctrl),model group(group M)and group PNS.Twelve rats in group M and group PNS were treated with liquid nitrogen freezing for femoral head necrosis.Three months later,rats in group M were injected with 2 mL of saline daily for two weeks,rats in group PNS were injected with 12.0 mg/mL of PNS saline solution 2 mL intraperitoneally,the group Ctrl gave no treatment.Rats in each group were executed 8 weeks later.Serum and femoral head specimens were collected for HE staining,Western Blotting for OPG(Osteoprotegerin),RANKL(Receptor Activator of NF-kB Ligand)and ELISA was used to determine the serum concentrations of PINP(Procollagen Type-I N Propeptide),TRAP-5b(Tartrate Resistant Acid Phosphatase-5b).Results:PNS reduced the empty lacunae rate of femoral head(P<0.01),down-regulated the expression of RANKL(P<0.01),up-regulate the expression of OPG(P<0.01),reduced the level of TRAP-5b(P<0.05)in serum,and increased the level of PINP in serum in the rats with ONFH.Conclusion:PNS can inhibit the activity of osteoclasts,promote osteogenesis and play a protective role in ONFH.
