摘要:
目的:观察活血通督汤对大鼠脊髓损伤后NLRP3,IL-1β及IL-18表达的影响,探讨该方对脊髓损伤后 炎症的作用及其机制。方法:将24只成年SD大鼠随机分为假手术组、模型组、活血通督汤组(各8只)。假手 术组行椎板切除术(不损伤脊髓),模型组和活血通督汤组采用NYU脊髓损伤打击器复制脊髓损伤模型。造模 成功后,假手术组和模型组给予生理盐水灌胃,活血通督汤组给予活血通督汤灌胃,给药剂量为12.6 mL/(kg ·d),术后第3天取材。采用免疫组化法检测NLRP3炎症体的表达,尼氏染色法观察神经细胞形态,ELISA法检 测炎症因子IL-1β和IL-18的表达。结果:尼氏染色显示活血通督汤组大鼠神经细胞的形态结构优于模型组 。免疫组化法显示:与假手术组比较,模型组NLRP3炎症体表达较高,差异有统计学意义(P<0.05); 与模型组 比较,活血通督汤组NLRP3表达减少,差异有统计学意义(P<0.05)。ELISA法检测结果显示模型组血清中IL-1 β和IL-18表达较假手术组高,差异有统计学意义(P<0.05); 活血通督汤组IL-1β和IL-18低于模型组。结 论:活血通督汤能抑制脊髓损伤后炎症反应,其机制可能与其抑制NLRP3的表达有关。
Abstract:
To observe the effect of Huoxue Tongdu decoction on the expression of NLRP3,IL-1β,IL-18 after spinal cord injury in rats,and to explore the mechanism of Huoxue Tongdu decoction on inflammation after spinal cord injury.Methods:24 adult SD rats were randomly divided into sham operation group,model group,Huoxue Tongdu decoction group (n=8).After the success of the model,the sham operation group and the model group were given normal saline every day.Huoxue Tongdu decoction group was given Huoxue Tongdu decoction.After 3 d,tissues were gathered for Nissl's staining,NLRP3 were detected by immunehistochemical staining while IL-1β,IL-18 were detected by ELISA.Results:Nissl's staining showed that the structure and morphology of neurons in the Huoxue Tongdu decoction group was better than that of model group with the improvement of the neuronal damage condition.Compared to sham group,there were more NLRP3 expression in the model group (P<0.05),and the expression of NLRP3 in Huoxue Tongdu decoction was less than that of model group(P<0.05).Compared to sham group,there were more IL-1β,IL-18 expression in the model group(P<0.05),and the expression of IL-1β,IL-18 in Huoxue Tongdu decoction was less than that of model group(P<0.05).Conclusion:Huoxue Tongdu decoction can inhibit inflammation and promote rehabilitation and repairment of neuron after acute spinal cord injury with a mechanism that Huoxue Tongdu decocotion can inhibit the expression of NLRP3.
