摘要:
目的:观察活血通督汤对坐骨神经损伤后脊髓损伤性反应的作用机制。方法:将16只SD大鼠切断双侧坐骨神经后,随机分为模型组(8只)和活血通督汤组(8只)。活血通督汤组给予活血通督汤治疗,连续4周,2次/d; 模型组灌服等量0.9%NaCl溶液,2次/d.4周后取出大鼠脊髓固定、包埋。应用免疫组织化学法检测脊髓中脑源性神经营养因子(Brain-derived Neurotrophic Factor,BDNF)和胶质原纤维酸性蛋白(Glial Fibrillary Acidic Protein,GFAP)的表达,并用图像分析软件检测表达量。结果:活血通督汤组BDNF的表达明显高于模型组(P<0.01),模型组GFAP的表达明显高于活血通督汤组(P<0.01)。结论:活血通督汤保护脊髓损伤性反应的作用在于促进BDNF的产生,也与抑制星形胶质细胞的活性有关。
Abstract:
Objective: To observe the effects of huoxuetongdu decoction on the injury reaction of spinal cord after sciatic nerve injury. Methods: All 16 SD rats were be cut off bilateral sciatic nerve, then they were randomly divided into normal saline group(n=8)and huoxuetongdu decoction group(n=8). The rats of huoxuetongdu decoction group were gavaged with huoxuetongdu decoction twice a day, while normal saline group were gavaged with normal saline twice a day, for 4 weeks. Then, the spinal cord of all rats was extracted, fixed and embedded. The immunohistochemistry method was used to detect the expression of BDNF and GFAP in spinal cord, while image analysis software was used to measure the content of the expression. Results: The expression of BDNF in huoxuetongdu decoction group was significantly higher than normal saline group(P<0.01), the expression of GFAP in normal saline group was significantly higher than the huoxuetongdu decoction group(P<0.01). Conclusion: Huoxuetongdu decoction can promote the production of BDNF, and inhibition of astrocytes activity, thus protecting the injury reaction of spinal cord after peripheral nerve injury.
